槲皮素抑制宫颈癌Siha细胞增殖和侵袭实验研究 被引量:9

Effect of quercetin on inhibiting proliferation and invasion of human cervical cancer Siha cells

摘要 目的探讨槲皮素对人宫颈癌Siha细胞生长、增殖、黏附、迁移和侵袭的影响。方法对数生长期人宫颈癌Siha细胞,分为对照组和20、40、80μmol/L槲皮素组,分别加入0、20、40、80μmol/L槲皮素进行处理;采用MTT法检测各组细胞增殖率,采用划痕试验检测各组细胞迁移距离和迁移率,采用Transwell小室检测各组侵袭细胞数和侵袭抑制率,采用实时定量PCR反应检测对照组与80μmol/L槲皮素组细胞HOTAIR、miR-23b、MAPK1mRNA表达水平,采用Western blot法检测各组细胞HOTAIR、miR-23b、MAPK1蛋白表达水平。结果对照组和20、40、80μmol/L槲皮素组细胞增殖率、细胞迁移距离、迁移率、侵袭细胞数依次降低(P<0.05),侵袭抑制率依次增高(P<0.05);80μmol/L槲皮素组细胞HOTAIR mRNA(0.723±0.089)、MAPK1 mRNA(0.786±0.166)表达水平低于对照组(1.665±0.104、1.637±0.151),miR-23bmRNA表达水平(6.671±0.093)高于对照组(1.578±0.112)(P<0.05);对照组和20、40、80μmol/L槲皮素组细胞HOTAIR蛋白(0.97±0.15、0.72±0.09、0.54±0.11、0.33±0.07)、MAPK1蛋白(0.74±0.07、0.42±0.08、0.21±0.05、0.13±0.03)表达水平依次降低,miR-23b蛋白表达水平(0.18±0.02、0.43±0.06、0.67±0.10、0.88±0.09)依次增高(P<0.05)。结论槲皮素明显抑制Siha细胞生长、增殖、迁移和侵袭,其机制可能与其下调长链非编码RNA HOTAIR并通过其靶向调控miR-23b/MAPK1轴有关。 Objective To investigate the influence of quercetin on the growth,proliferation,adhesion,migration and invasion of human cervical cancer Siha cells.Methods Human cervical cancer Siha cells in logarithmic growth phase were divided into control group and 20,40 and 80μmol/L quercetin groups,treated with 0,20,40 and 80μmol/L quercetin,respectively.The cell proliferation rate was detected by MTT,the cell migration distance and migration rate were detected by scratch test,and the cells count of invasion and invasion rate were detected by Transwell chamber in all groups.The expression levels of HOTAIR,miR-23 band MAPK1 mRNA were detected by real-time quantitative PCR in control group and 80μmol/L quercetin group.The expression levels of HOTAIR,miR-23 band MAPK1 proteins were detected by Western blot in all groups.Results The proliferation rate,cell migration distance,migration rate and count of invasion of Siha cells decreased gradually in control group and 20,40 and 80μmol/L quercetin groups in turns,and the proliferation inhibition rate increased gradually in turns(P<0.05).The mRNA expressions of HOTAIR and MAPK1 were significantly lower,and the mRNA expression of miR-23 bwas significantly higher in 80μmol/L quercetin group(0.723±0.089,0.786±0.166,6.671±0.093)than those in control group(1.665±0.104,1.637±0.151,1.578±0.112)(P<0.05).The expressions of HOTAIR protein(0.97±0.15,0.72±0.09,0.54±0.11,0.33±0.07)and MAPK1 protein(0.74±0.07,0.42±0.08,0.21±0.05,0.13±0.03)decreased gradually,and the expression of miR-23 bprotein(0.18±0.02,0.43±0.06,0.67±0.10,0.88±0.09)increased gradually in control group and 20,40 and 80μmol/L quercetin groups in turns(P<0.05).Conclusion Quercetin inhibits significantly the growth,proliferation,migration and invasion of Siha cells,which might be correlated with the down-regulation of long non-coding RNA HOTAIR and target regulation of miR-23 b/MAPK1 axis.
