摘要
为了研究[60Co]γ射线诱导小麦Glu-1A等位基因突变体的品质差异与遗传规律,用300 Gy的[60Co]γ射线辐射皖麦50的干种子,用近红外品质测定和SDS-PAGE法,对获得的Glu-1A等位基因品质量差异进行了评价和遗传规律研究,结果表明:在M2筛选到一个HMW-GS突变体,Glu-1ANull突变为Glu-1A1,突变率为0.15%。突变体与皖麦50杂交,在F2品质性状有超亲现象,品质得分与吸水率和硬度之间相关显著。在F1的正、反交,HMW-GS表现为双显性的Glu-1A1,侧交的Glu-1A1∶Glu-1ANull符合1∶1的分类规律,F2的Glu-1A1∶Glu-1ANull符合3∶1的分类规律。突变体蛋白质含量增加,杂交后代有品质超亲现象,遗传表现符合孟德尔独立遗传和自由组合规律。
Inorder to study quality differences and inheritance of Glu-1A Alleles mutant of wheat induced by[60Co]γ-ray,with 300 Gy of [60Co]γ-ray radiation of dry wheat seeds of Wanmai 50,and with method of analysis of near infrared and SDS-PAGE,quality evaluation and differences was made and inheritance of Glu-1A Alleles mutant was studied. The results showed that one HMW-GS mutant was screened in M2,Glu-1A subunit component changed from Glu-1ANull to Glu-1A1,and the mutation ratio was 0. 15%. The quality traits of the mutant Wanmai50 in F2 had transgressive phenomena. There was significant correlation between quality score and water absorption,hardness and sedimentation. The HMW-GS of orthogonal and reciprocal cross of F1 all showed double dominance of Glu-A1. The ratio of Glu-1A1 to Glu-1ANull was in line with 1 ∶ 1 segregation rule in side-crosses,and was in line with 3 ∶ 1 in F2. In conclusion,the protein content of mutant increased,the hybrids had super-pro-quality,and the inheritance accorded with the Mendelian inheritance and free combination.
