摘要 目的 研究白藜芦醇 (Res)对小鼠移植性肝癌组织中细胞周期蛋白CyclinD1,CyclinB1和p34cdc2表达的影响 ,探讨Res抗肝癌作用及其机制 .方法 利用小鼠移植性肝癌H2 2模型研究Res的体内抗肿瘤效果 ,免疫组织化学检测实验组与对照组小鼠肝癌及瘤旁肝组织CyclinD1,CyclinB1和p34cdc2蛋白的表达情况 .结果 Res治疗组能不同程度抑制小鼠肝脏移植瘤的生长 ,10mg·kg- 1 和 15mg·kg- 1 的Res治疗时抑瘤率分别为 36 .3%和 4 9.3% ,与对照组比较有显著差异 (P <0 .0 5 ) .与对照组相比 ,Res治疗组癌组织中CyclinD1的表达无明显变化 (6 6 .7%vs 6 0 .0 % ,P >0 .0 5 ) ,而CyclinB1(77.8%vs30 .0 % ,P <0 .0 5 )和p34cdc2 (88.9%vs 4 0 .0 % ,P <0 .0 5 )的表达明显降低 .结论 Res具有抗肝癌作用 .通过抑制Cy clinB1和p34cdc2的表达影响细胞周期的进程 ,可能是Res抗癌作用的机制之一 . AIM To explore the effect of resveratrol on the expression of cell cycle proteins including CyclinD1, CyclinB1 and p34cdc2 in murine transplantable liver cancer; to discuss the antitumor activity of resveratrol and its mechanism. METHODS Transplantable murine hepatoma22 model was used to evaluate the antitumor activity of resveratrol in vivo ; the protein expression of CyclinD1, CyclinB1 and p34cdc2 gene in the tumor and adjacent noncancerous liver tissues was measured by immunohistochemistry methods. RESULTS Resveratrol could inhibit the growth of murine transplantable liver cancer, after the mice bearing H22 tumor were treated with 10 or 15 mg·kg -1 resveratrol for 10 days, the inhibition rate are 36.3% and 49.3%, respectively, increased obviously compared with control group( P <0.05). Depressed expression of CyclinB1, p34cdc2 protein was found in the transplantable murine hepatoma22 treated with resveratrol, but the expression of CyclinD1 protein has not changed. CONCLUSION Resveratrol has obvious antitumor activities on murine hepatoma22. Resveratrol can inhibit the progression of cell cycle by decreasing the expression of CyclinB1, p34cdc2 protein and may be one of its antitumor mechanism.
