摘要
本研究旨在优化超高效液相色谱(UPLC)方法,以测定葡萄和葡萄酒中的7种花色苷。通过比较Shimpack GIST C_(18)色谱柱(2.1 mm×100 mm,2μm)和Kinetex PFP色谱柱(3 mm×100 mm,2.7μm)的分离效果,最终选择Kinetex PFP色谱柱进行后续试验。葡萄样品采用甲酸∶甲醇(2∶98,V/V)进行提取,葡萄酒样品直接进样。流动相A为甲酸水溶液,体积比为3∶97,流动相B为甲酸乙腈溶液,体积比为2∶98。试验采用梯度洗脱模式进行分离,检测波长设定为518 nm,进样量为1μL。在优化的色谱条件下,7种花色苷在11 min内实现了良好的分离效果,其浓度与峰面积之间呈现出良好的线性关系(R^(2)>0.9981)。回收率介于87.3%~92.91%,精密度(RSD)低于1%。该方法能够快速、准确地对葡萄及葡萄酒中的花色苷进行定性与定量分析,具备较高的可靠性和应用价值。
The objective of the present study was to optimise an ultra performance liquid chromatography(UPLC)method for the determination of seven anthocyanins in grapes and wines.Following a rigorous comparison of the separation effects exhibited by the Shim-pack GIST C_(18) column(2.1 mm×100 mm,2μm)and the Kinetex PFP column(3 mm×100 mm,2.7μm),it was determined that the latter was to be selected as the stationary phase.The grape samples were extracted using formic acid:methanol(2∶98,V/V),and the wine samples were injected directly.The mobile phase A was composed of formic acid in water,with a volume ratio of 3∶97,while mobile phase B was a mixture of formic acid and acetonitrile,with a volume ratio of 2∶98.The separation process was conducted in gradient elution mode,with a detection wavelength of 518 nm and an injection volume of 1μL.It was demonstrated that under optimised chromatographic conditions,the seven anthocyanosides were successfully separated within 11 minutes.Furthermore,an excellent linear relationship was observed between the concentration and the peak area(R^(2)>0.9981).The range of recoveries was from 87.3%to 92.91%,with a precision(RSD)value below 1%.The method has been demonstrated to facilitate rapid and accurate qualitative and quantitative analysis of anthocyanosides in grapes and wines.It is characterised by high reliability and significant application value.
