5种澳洲坚果种质材料叶片对低温胁迫的生理响应

摘要 【目的】探明不同澳洲坚果种质对低温胁迫的响应和耐受性,为培育贵州本地适应性广、耐寒性强的澳洲坚果种质提供参考。【方法】以澳洲坚果良种O.C和GR1为对照,采用培养箱在-4℃下对5种澳洲坚果种质材料的叶片进行低温处理,并对10个耐寒生理指标进行测定,分析不同澳洲坚果种质材料叶片对低温胁迫的响应,并利用隶属函数法对5个澳洲坚果种质材料的耐寒性进行综合评价。【结果】低温胁迫下,澳洲坚果不同材料的各生理指标发生显著变化,各材料的MDA含量、脯氨酸含量、可溶性糖含量、可溶性蛋白含量、相对电导率、SOD活性、POD活性、CAT活性、PPO活性显著升高,相对含水量降低。随低温胁迫时间的延长,各材料的耐寒性表现存在差异。QA1在MDA含量、脯氨酸含量、可溶性蛋白含量方面的表现优于O.C和GR1,其在相对电导率方面的表现优于GR1;QA2在MDA含量、POD活性、PPO活性方面的表现优于O.C和GR1,其在相对电导率、CAT活性、可溶性蛋白含量方面的表现优于GR1,在脯氨酸含量方面的表现与O.C和GR1相当;QA3在MDA含量方面的表现优于O.C,在可溶性蛋白含量、相对电导率、相对含水量方面的表现优于GR1,在SOD活性方面的表现优于O.C和GR1。此外,与QA1相比,QA2和QA3表现出较全面的耐寒性。【结论】低温胁迫下,黔澳系列澳洲坚果通过不同的生理调节机制的协同作用来应对低温胁迫,以提高自身的耐寒能力。隶属函数法分析结果表明,5种澳洲坚果种质按照耐寒性由强到弱排序依次为O.C、GR1、QA3、QA2、QA1。 【Objective】In order to explore the response and tolerance of different macadamia germplasm to low temperature stress,it provided a theoretical basis for breeding local macadamia germplasm with wide adaptability and strong cold tolerance in Guizhou.【Method】The leaves of 5 macadamia germplasm materials were treated at-4℃in an incubator with the control of O.C and GR1,and 10 physiological indexes of cold tolerance were determined to analyze the response of leaves of different Macadamia germplasm materials to low temperature stress,and the cold hardiness of the five macadamia germplasm materials was comprehensively evaluated by the method of the affiliation function.【Result】Under low temperature stress,the physiological indexes of different macadamia materials changed significantly.The contents of MDA,proline,soluble sugar,soluble protein,relative electrical conductivity and the activities of SOD,POD,CAT and PPO of each material increased significantly,and the relative water content decreased.With the prolongation of low temperature stress,there were differences in the cold hardiness of each material.QA1 was better than O.C and GR1 in MDA,proline,soluble protein,and better than GR1 in relative electrical conductivity.QA2 was better than O.C and GR1 in MDA,POD In MDA,POD and PPO,QA2 performed better than O.C,better than GR1 in MDA,relative electrical conductivity,POD,CAT,PPO and soluble protein,and comparable to GR1 and O.C in Pro.QA3 performed better than O.C in MDA and SOD,and better than GR1 in SOD,soluble protein,relative electrical conductivity and relative water content.In addition,compared with QA1,QA2 and QA3 showed more comprehensive cold tolerance.【Conclusion】Under low temperature stress,Qian’ao series of macadamia nuts respond to low temperature stress through different physiological regulation mechanisms to improve their cold tolerance.By membership function analysis,the order of cold tolerance of 5 Macadamia nut germplasm was O.C,GR1,QA3,QA2,QA1.
