摘要
旨在探究1-棕榈酰-2-油酰-sn-甘油-3-磷酸胆碱(POPC)对绵羊卵母细胞体外成熟和体外受精的影响,探索POPC的最佳添加剂量,以优化体外胚胎生产技术。通过免疫荧光染色、体外成熟培养和体外发育试验,研究POPC对绵羊卵母细胞体外成熟和体外受精的影响,将POPC以不同浓度添加至IVM培养基中,观察卵丘细胞扩张程度、第一极体排出率、ROS水平以及体外受精后的卵裂率和囊胚率。结果显示,添加150或200μg/mL的POPC可以促进卵丘细胞扩张,提高卵母细胞体外成熟效率;加入100μg/mL的POPC能极显著提高绵羊卵母细胞的第一极体排出率,并且极显著降低ROS水平,而加入150μg/mL的POPC则能极显著提升其受精后的发育效率。
This study aimed to explore the effect of POPC on in vitro maturation and in vitro fertilization of sheep oocytes,and to explore the optimal additive dose of POPC to optimize the in vitro embryo production technology.The effects of POPC on in vitro maturation and in vitro fertilization of sheep oocytes were studied by immunofluorescence staining,in vitro maturation culture and in vitro development test,the expansion degree of cumulus cells,the first polar body expulsion rate,ROS level,cleavage rate and blastocyst rate after in vitro fertilization were observed.The results showed that 150 and 200μg/mL POPC could promote cumulus cell expansion and improve the efficiency of oocyte maturation in vitro The addition of 100μg/mL POPC could extremely significantly increase the first polar body expulsion rate of sheep oocytes and extremely significantly reduce the ROS level,while the addition of 150μg/mL POPC could extremely significantly increase the development efficiency after fertilization.
