山新杨PdaDREB2E基因的克隆及表达模式分析

Cloning and Expression Pattern Analysis of PdaDREB2E Gene in Populus davidiana×P.alba var.pyramidalis

摘要 为鉴定山新杨中响应非生物胁迫的DREB基因,从山新杨中克隆得到1条PdaDREB2E基因,利用生物信息学分析对该基因氨基酸序列特征进行分析,结果表明,PdaDREB2E蛋白序列共有451个氨基酸,相对分子质量为50334.94,理论等电点(pI)为6,不存在跨膜区及信号肽,为非分泌蛋白,包含31个丝氨酸(S),11个苏氨酸(T),5个酪氨酸(Y)磷酸化位点。二、三级结构中主要由无规卷曲(41.68%),α-螺旋(34.37%),延伸链(14.19%),β-折叠(9.76%)组成。该基因定位于细胞核,具有自激活活性。系统进化树分析表明,该氨基酸序列与银白杨和毛白杨中DREB2E蛋白的亲缘关系较近。利用实时荧光定量PCR(RT-qPCR)技术对该基因在不同组织和非生物胁迫下的表达模式进行分析,结果显示PdaDREB2E基因在根中的表达量最高,茎中最低;PdaDREB2E基因在响应干旱(PEG)、盐(NaCl)非生物胁迫及脱落酸(ABA)处理中呈现出不同的表达特点。该结果为研究山新杨PdaDREB2E基因的抗逆功能研究提供材料。 To identify the DREB genes in Populus davidiana×P.alba var.pyramidalis responding to abiotic stress,a PdaDREB2E gene was cloned from Populus davidiana×P.alba var.pyramidalis in this study.The amino acid sequence characteristics of this gene were analyzed by bioinformatics.The results showed that the protein sequence of PdaDREB2E contained 451 amino acids,with a relative molecular mass of 50334.94 and a theoretical isoelectric point(pI)of 6.There were no transmembrane regions or signal peptides,indicating that it was a non-secreted protein.It contained 31 serine(S),11 threonine(T),and 5 tyrosine(Y)phosphorylation sites.The secondary and tertiary structures were mainly composed of random coils(41.68%),α-helices(34.37%),extended chains(14.19%),andβ-sheets(9.76%).The gene was located in the nucleus and had self-activation activity.Phylogenetic tree analysis indicated that the amino acid sequence was closely related to the DREB2E proteins in Populus alba and Populus tomentosa.The expression pattern of the gene in different tissues and under abiotic stress was analyzed by real-time fluorescence quantitative PCR(RT-qPCR).The results showed that the expression level of PdaDREB2E was the highest in roots and the lowest in stems.Under abiotic stress such as drought(PEG),salt(NaCl),and abscisic acid(ABA)treatment,PdaDREB2E showed different expression characteristics.The results provided materials for the study of the stress resistance function of the PdaDREB2E gene in Populus davidiana×P.alba var.pyramidalis.
