不同剂量促熟激素对欧洲鳗鲡卵巢成熟过程中性激素变化的影响

The effect of different doses of ripening hormones on the changes of sex hormones during the ovarian maturation process of Anguilla anguilla

摘要 为评估人工促熟过程中不同剂量促熟激素对欧洲鳗鲡(Anguilla anguilla)卵巢中性激素分泌的影响。实验使用鲤鱼垂体(CPE)和人绒毛膜促性腺激素(HCG)混合匀浆作为促熟激素(CPE:6.5 mg,HCG:200 IU)/mL,设置1.5,1,0.5 mL/kg三种注射剂量组,采用酶联免疫吸附法测定促卵泡激素(FSH)、促黄体生成激素(LH)、雌二醇(E_(2))、睾酮(T)、11-酮基睾酮(11-KT)和17α,20β-二羟基-4-孕烯-3-酮(DHP)六种性激素在卵巢中的含量变化。结果显示,促熟激素处理显著影响了欧洲鳗鲡卵巢发育过程中的性激素水平,其中1.5 mL/kg剂量组对6种性激素水平的调节效果最为显著,在注射16针促熟激素后卵巢发育成熟,六种性激素含量分别为:FSH(7.14±0.05)U/g、LH(14061.6±179.53)pg/g、E_(2)(22.09±0.15)ng/g、T(2.01±0.01)nmol/g、11-KT(1722.78±6.80)pg/g、DHP(13.51±0.09)ng/g,其中FSH的含量变化与T和11-KT趋势基本一致,LH含量变化与E_(2)、DHP趋势基本一致。本研究表明,注射1.5 mL/kg促熟激素在欧洲鳗鲡人工促熟过程中能显著促进卵巢关键性激素(FSH、LH、E_(2)、T、11-KT、DHP)合成并在第12和16次注射后达到含量峰值。 The Anguilla anguilla currently faces severe challenges posed by the continuous decline of germplasm resources and soaring market prices,which have become major bottlenecks restricting the sustainable development of its aquaculture industry.The breakthrough in artificial breeding technology represents a core solution to address the shortage of fry.However,the complex reproductive biology of the A.anguilla has thus far hindered the commercial application of artificial breeding techniques.Key challenges include the instability of exogenous hormones for inducing maturation,asynchronous gonadal development among broodstock,and poor sperm quality,which collectively contribute to the limited research on the effects of hormones and their optimal dosages for promoting eel maturation during total artificial reproduction.In this study,different dosages of maturation-promoting hormones were systematically evaluated for their impacts on the changes in sex body index and key sex hormone concentrations during ovarian development in A.anguilla.Three dosage groups were established:Group A(1.5mL/kg),Group B(1mL/kg),and Group C(0.5mL/kg).Enzyme-Linked Immunosorbent Assay was employed to quantify ovarian concentrations of six reproductive hormones:follicle-stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E_(2)),testosterone(T),11-ketotestosterone(11-KT),and 17α,20β-dihydroxy-4-pregnen-3-one(DHP).The results showed that hormonal treatment significantly modulated sex hormone levels during ovarian development.The 1.5mL/kg do-sage group exhibited the most pronounced regulatory effects,achieving gonadal maturation after 16 consecutive injections.Final hormone concentrations in this group were quantified as follows:FSH(7.14±0.05)U/g,LH(14,061.6±179.53)pg/g,E_(2)(22.09±0.15)ng/g,T(2.01±0.01)nmol/g,11-KT(1722.78±6.80)pg/g,and DHP(13.51±0.09)ng/g.Notably,the changes in the content of FSH followed the same trend as that of T and 11-KT,while the changes in the content of LH were consistent with the trend of E_(2) and DHP.This study showed that injecting 1.5 ml of maturation-inducing hormone could significantly promote the key ovarian hormones(FSH,LH,E_(2),T,11-KT,DHP)during the artificial maturation process of A.anguilla reaching peak levels after the 12th and 16th injections.The results provided a robust scientific foundation for achieving efficient regulation of A.anguilla maturation,facilitating high-quality gonadal development,and precisely manipulating maturation and spawning processes.These insights are pivotal for advancing the full artificial reproduction of the A.anguilla,addressing critical bottlenecks in aquaculture sustainability and conservation efforts.
