毛头鬼伞多糖基于生物钟调控对短期睡眠限制小鼠糖脂代谢的影响

Effects of Coprinus comatus polysaccharides on glucose and lipid metabolism in short-term sleep restriction mice based on biological clock regulation

摘要 构建C57BL/6小鼠短期睡眠限制(SR)模型以及脑和肌肉组织芳香烃受体核转运蛋白样1(Bmal1)基因敲除SR小鼠模型,分析毛头鬼伞多糖(CCP)对SR小鼠葡萄糖耐量、胰岛素耐量、血液生化指标、血脂以及糖脂代谢相关基因、蛋白质和生物时钟基因(Clock)/Bmal1信号通路的影响。结果表明:CCP能显著改善SR小鼠的葡萄糖耐量和胰岛素耐量,降低血糖、血脂水平,同时影响参与糖脂代谢的过氧化物酶体增殖物激活受体α(PPARα)、脂肪酸合酶(FASN)、脂蛋白脂肪酶(LPL)和固醇调节元件结合蛋白1(SREBP-1)基因的表达水平。其机制与Clock/Bmal1信号通路的Clock、Bmal1表达水平有关。 Short-term sleep restriction(SR)models of C57BL/6 mice and Bmal1 knockout SR mouse models were constructed to analyze the effects of Coprinus comatus polysaccharide(CCP)on glucose tol-erance,insulin tolerance,blood biochemical parameters,blood lipids,as well as genes,proteins related to glucose and lipid metabolism,and the Clock/Bmall signaling pathway in SR mice.The results showed that CCP significantly improved glucose tolerance and insulin tolerance in SR mice,reduced blood glu-cose and lipid levels,and affected the expression levels of genes involved in glucose and lipid metabo-lism,including peroxisome proliferator-activated receptorα(PPARa),fatty acid synthase(FASN),lipo-protein lipase(LPL),and sterol regulatory element-binding protein 1(SREBP-1).The mechanism was related to the regulation of Clock and Bmall expression levels in the circadian clock gene(Clock)/aryl hydrocarbon receptor nuclear translocator-like protein 1(Bmall)signaling pathway.
