超高压辅助低共熔溶剂提取地黄多糖工艺优化及其抗氧化活性研究

Study on optimization of ultra-high pressure assisted deep eutectic solvent extraction process of Rehmannia glutinosa polysaccharides and their antioxidant activity

摘要 采用超高压辅助低共熔溶剂(deep eutectic solvent, DES)法提取地黄多糖,在筛选出最佳DES的基础上,通过单因素试验和响应面试验优化地黄多糖提取工艺,并评价地黄多糖对DPPH·、·OH的清除作用。结果表明:最佳提取工艺条件为DES含水量31%、提取压力403 MPa、提取时间8 min,在此条件下地黄多糖得率为8.27%。制备的地黄多糖具有较高的DPPH·、·OH清除能力。表明超高压辅助DES提取工艺是制备高得率、高抗氧化活性地黄多糖的适宜方法。 The ultra-high pressure assisted deep eutectic solvent(DES)method was used to extract poly-saccharides from Rehmannia glutinosa.Based on the selection of the optimal DES,the extraction process of Rehmannia glutinosa polysaccharides was optimized through single factor experiments and response sur-face experiments,and the effects of Rehmannia glutinosa polysaccharides on DPPH·,·OH scaven-ging ability were evaluated.The results showed that the optimal extraction process conditions were DES moisture content 31%,extraction pressure 403 MPa,and extraction time 8 min.Under these condi-tions,the yield of Rehmannia glutinosa polysaccharides was 8.27%.The prepared Rehmannia glutinosa polysaccharides had high DPPH,"OH scavenging ability,which indicated that the ultra-high pres-sure assisted DES extraction process was a suitable method for preparing high-yield and high antioxidant activity Rehmannia glutinosa polysaccharides.
