摘要
研究海南岛海域野生花鳗鲡(Anguilla marmorata)线粒体基因组序列,有助于理解不同鳗鲡属物种之间的亲缘关系,并为鳗鲡属鱼类的鉴定和系统进化提供科学依据。采用Illumina技术对2021年11月采自海南省陵水黎族自治县田新村加薪河的10尾花鳗鲡样品线粒体基因组进行测序,结果显示线粒体基因组全长为16712 bp,碱基组成为A(34.30%)、C(25.93%)、T(24.57%)和G(15.20%);A+T含量为58.87%,呈现出明显的AT偏好性。线粒体基因组结构包含37个基因和1个控制区(D-loop)。对花鳗鲡13个蛋白质编码基因(protein-coding genes,PCGs)密码子的相对同义密码子使用度(RSCU)进行统计分析,发现除AGC和ATC外,其余密码子的RSCU均大于1,表明均为偏好性密码子;比较中国和印度尼西亚海域的花鳗鲡线粒体基因组,发现除COI和ND6基因外,其他PCGs的相似性均高于99%;进一步对同属的日本鳗鲡(A.japonica)、吕宋鳗鲡(A.luzonensis)、双色鳗鲡(A.bicolor)和云纹鳗鲡(A.nebulosa)线粒体基因组的13个PCGs进行两两比对分析,结果显示这5种鳗鲡属鱼类的PCGs的相似度为90%~97%。通过分析23种鳗鲡目鱼类线粒体基因组中13个PCGs的核苷酸序列,构建了系统进化树,成功地将花鳗鲡与其他鳗鲡属鱼类区分开来。研究结果表明花鳗鲡与内唇鳗鲡(A.interioris)同属一个分支,二者亲缘关系最近。
In this study,we analyzed the mitochondrial genome structure of Anguilla marmorata(giant mottled eel)on Hainan Island and explored the phylogenetic relationships of A.marmorata with other Anguilliformes species by comparing their mitochondrial genome structures.We aimed to provide a scien-tific foundation for species identification,evolutionary analysis,environmental DNA(eDNA)quantifi-cation,and the conservation and restoration of wild populations of this species.In November 2021,10 A.marmorata samples were collected in Hainan Island for DNA extraction and mitochondrial genome se-quencing using Illumina technology.The complete mitochondrial genome has a total length of 16712 bp,with a base composition of A(34.30%),C(25.93%),T(24.58%),and G(15.20%).The A+T content is 58.87%,demonstrating a pronounced AT bias.The mitochondrial genome of A.marmorata is comprised of 37 genes and one control region(D-loop).Statistical analysis of the Relative Synonymous Codon Usage(RSCU)for the 13 protein-coding genes(PCGs)in A.marmorata revealed that all codons,except of AGC and ATC,exhibited RSCU values exceeding 1,indicating strong codon usage bias in favor of these codons.Comparative analysis of mitochondrial genomes among A.marmorata populations from Chinese and Indonesian waters revealed all PCGs exhibited sequence similarity exceeding 99%except for the COI and ND6 genes.Pairwise analysis of the 13 PCGs in the mitochondrial genomes of A.marmorata,A.japonica,A.luzonensis,A.bicolor and A.nebulosa had a PCG sequence similarity from 90%to 97%among the five Anguillas species.Phylogenetic analysis of 23 Anguilliforme species based on nucleotide sequences of the 13 PCGs in the mitochondrial genomes successfully distinguished A.marmorata from other Anguilla species.The results revealed that A.marmorata and A.interioris form a monophyletic clade.This study provides comprehensive insights into the complete mitochondrial genome sequence of wild A.marmorata in Hainan Island,contributing valuable information for understanding inter-species re-lationships within Anguilla species and offering support for the identification and phyloevolutionary stud-ies of Anguilla.
