摘要
对已克隆化分离的一个β地贫基因,应用6种人工合成的寡核苷酸为内引物。以[α-^(35)S]dATP进行标记,用Sanger双脱氧末端终止法进行序列测定。结果表明,此例β珠蛋白基因的密码子41/42中缺失了4个核苷酸(TCTT)。
DNA sequence of βg-lobin gene from a patient with p-thalassemia major in Guangdong province was subcloned into Ml3mp 19 and analyzed by dideoxy-chain termination method of Sanger,using six 19mer oligonu-cleotides as internal primers and (α-^(35)S) dATP for labeling in synthetic mixture.A four-nucleotide deletion in codon 41 and 42 was found.This is the firstβ-thalassemia gene cloned and sequenced in China.
