响应面法优化花椒挥发油微囊制备工艺

Optimize preparation technology of essential oil from Zanthoxylum bungeanum Maxim microencapsule with response surface analysis

摘要 试验旨在优化花椒挥发油微囊的制备工艺。以明胶和海藻酸钠为壁材,采用复凝聚法制备花椒挥发油微囊,通过单因素试验探究明胶和海藻酸钠的比例、反应pH值、壁材浓度、乳化剂用量和搅拌转速等因素对微囊形成的影响。以微囊的包封率为评价指标,利用响应面法筛选出制备花椒挥发油微囊的最优条件。结果显示,明胶海藻酸钠比例为4∶1,反应pH值为4.55,壁材浓度为1.773%,乳化剂用量为1.27%,搅拌转速为743 r/min时,制备出的花椒挥发油微囊包封率最高,为88.13%,且微囊能够缓释花椒挥发油。研究表明,试验使用复凝聚法制备微囊化包封率高,缓释效果良好,可为未来开发新型挥发油制剂治疗奶牛乳房炎提供参考。 The experiment aimed to optimize the preparation process of Zanthoxylum bungeanum Maxim essential oil microencapsule.Using gelatin and sodium alginate as wall materials,Zanthoxylum bungeanum Maxim essential oil microencapsule is prepared by the complex coacervation method.Single-factor experiments were conducted to investigate the effects of the gelatin-to-sodium alginate ratio,reaction pH value,wall material concentration,emulsifier dosage,and stirring speed on microencapsule formation.The encapsulation efficiency of the microencapsule was used as the evaluation index,and response surface methodology was employed to determine the optimal preparation conditions for the Zanthoxylum bungeanum Maxim essential oil microencapsule.The results showed that when the ratio of gelatin-to-sodium alginate was 4∶1,the reaction pH was 4.55,the wall material concentration was 1.773%,the emulsifier dosage was 1.27%,and the stirring speed was 743 r/min,the encapsulation efficiency of the prepared Zanthoxylum bungeanum Maxim essential oil microencapsule was the highest,reaching 88.13%,and the essential oil microencapsule could achieve sustained release of Zanthoxylum bungeanum Maxim essential oil.The study indicates that the complex coacervation method used in the experiment yields microencapsule with high encapsulation efficiency and excellent sustained-release properties.This process provides reference for the future development of novel essential oil formulations for the treatment of bovine mastitis.
