营养条件与四环素浓度对大肠杆菌生长与突变的联合影响

摘要 为探究不同营养条件下抗生素对大肠杆菌的胁迫效应,设计了基于Luria-Bertani(LB)和Mueller-Hinton(MH)这2种培养基营养条件下四环素对大肠杆菌的生长和突变实验,并通过交互效应值判别培养基条件调整与四环素胁迫对其的联合作用模式。结果表明,在LB和MH培养基中,四环素对大肠杆菌生长表现出抑制作用,且随着培养基浓度增加,EC_(50)降低,毒性效应增强,此外在低浓度MH培养基中表现出低浓度促进、高浓度抑制的Hormesis效应。与MH培养基相比,LB培养基中四环素(tetracycline hydrochloride,TH)对E.coli的毒性作用更为显著。且随着培养基浓度增加,EC_(50)逐渐减小,TH对大肠杆菌的毒性逐渐增强。TH在LB和MH培养基中诱导大肠杆菌的突变频率促进率表现为先增大再减小最后转为抑制作用,其最大值分别为11.1%~32.3%和14.9%~313.6%,表明营养成分丰富度减少有利于大肠杆菌突变。培养基浓度降低和四环素胁迫对抑制大肠杆菌生长、促进突变的联合作用模式分别表现为拮抗、协同,相反地,浓度增加则分别表现为协同、拮抗。综上,基于低营养条件下的四环素虽然对大肠杆菌的毒性降低,却产生了更大的突变效应,这可能导致环境中大肠杆菌耐药性加剧,进而危害人体健康与生态系统安全,所以建议将其纳入抗生素风险评估的考量要素之中。 To investigate the stress effects of antibiotics on Escherichia coli under diverse nutritional conditions,growth and mutation experiments of tetracycline on Escherichia coli were conducted in two types of culture media with distinct nutritional profiles:Luria-Bertani and Mueller-Hinton.The interaction effect value was employed to determine the combined effect mode of the culture media condition and the tetracycline stress.The results showed that tetracycline exhibited inhibitory effects on the growth of Escherichia coli in Luria-Bertani and Mueller-Hinton,and as the concentration of the culture media increased,the EC_(50) decreased and the toxic effects increased.In addition,Mueller-Hinton with low concentrations showed a Hormesis effect of low concentration promoting and high concentration inhibiting.Compared with Mueller-Hinton,the toxic effect of tetracycline on Escherichia coli in Luria-Bertani was more significant.Moreover,as the medium concentration increased,the EC_(50) gradually decreased,and the toxicity of tetracycline on Escherichia coli gradually increased.The promotion rate of tetracycline-induced mutation frequency of Escherichia coli in Luria-Bertani and Mueller-Hinton showed an increase followed by a decrease,and finally turned into an inhibitory effect.The maximum values were 11.1%-32.3%and 14.9%-313.6%,respectively,demonstrating that reducing nutritional richness was beneficial for Escherichia coli mutations.The combined effect of culture media concentration reduction and tetracycline stress on inhibiting bacterial growth was generally antagonistic,while on promoting bacterial mutation was generally synergistic.On the contrary,the combined effect of culture media concentration increase and tetracycline stress was generally synergistic on inhibiting Escherichia coli growth and antagonistic on promoting Escherichia coli mutation.In summary,tetracycline induced greater bacterial mutagenic effects despite lower toxicity to Escherichia coli under low nutrient concentrations,which might lead to increased resistance of Escherichia coli in the environment,thereby endangering human health and ecosystem safety.Therefore,it was recommended that nutritional conditions were taken into account in a ntibiotic risk assessment.
