摘要
以艾草为研究对象,通过正交试验优化蒸馏时间、料液比(艾草∶水)和浸泡时间对精油得率的影响。采用羟基自由基清除法、1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除法及2,2′-联氮-双(3-乙基苯并噻唑啉-6-磺酸)(ABTS)自由基清除法评价艾草精油的抗氧化活性;通过滤纸片法、双倍稀释法等评估艾草精油的抑菌活性。结果表明:艾草精油的最佳提取条件为蒸馏时间1.5h、料液比1∶10(质量比)、浸泡时间2.0h。艾草精油对羟基自由基、DPPH及ABTS自由基的清除率随浓度升高而增强,最高分别达64.25%、50.23%和70.45%。此外,艾草精油对金黄色葡萄球菌和大肠杆菌均有抑菌作用,抑菌圈直径分别为24.00mm和19.67mm;对金黄色葡萄球菌的最小抑菌浓度(MIC)为37.5μL/mL,最小杀菌浓度(MBC)为75μL/mL;对大肠杆菌的MIC为75μL/mL,MBC为150μL/mL,且在2×MIC浓度下可完全抑制细菌生长。
The study investigated mugwort(Artemisia argyi)essential oil,optimizing the extraction process via orthogonal experiments to evaluate the effects of distillation time,solid-liquid ratio(mugwort:water),and soaking time on oil yield.Antioxidant activity was assessed using hydroxyl radical scavenging,l,l-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging,and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radical scavenging assays.Antibacterial activity was evaluated via filter paper diffusion and double dilution methods.Results showed that the optimal extraction conditions are distillation time of 1.5h,solid-liquid ratio of 1:10,and soaking time of 2.Oh.The scavenging rates of hydroxyl,DPPH,and ABTS radicals increased with concentration,reaching 64.25%,50.23%,and 70.45%respectively.Mugwort essential oil inhibits Staphylococcus aureus and Escherichia coli,with inhibition zones of 24.00mm and 19.67mm,respectively.The minimum inhibitory concentrations(MICs)were 37.5μL/mL for S.aureus and 75μL/mL for E.coli;the minimum bactericidal concentrations(MBCs)were 75μL/mL and 150μL/mL respectively.Bacterial growth was completely inhibited at 2×MIC.
