摘要
目的 探讨黑醋栗改善2型糖尿病(T2DM)大鼠心功能、心肌肥厚的作用机制。方法 将40只成年雄性SD大鼠随机分为4组:正常对照组、模型组(建立T2DM模型)、阳性对照组(建模+给予皮下注射利拉鲁肽)、黑醋栗组(建模+给予黑醋栗水煎剂)。建模10周时观察大鼠行为学变化,测定其体重及进行糖耐量实验,采用超声检测大鼠心功能,采用全自动生化仪检测血常规、血脂指标、肝肾功能、心肌酶指标,RT-PCR检测大鼠心肌组织中高迁移率蛋白1(HMGB1)、核因子-kappa B(NF-κB)m RNA的表达。结果 行为学观察显示,正常对照组大鼠毛发光泽,反应较为灵敏;模型组大鼠毛发干枯,出现多饮、多食、多尿症状,活动能力下降;阳性对照组和黑醋栗组大鼠毛发干枯、多饮、多食、多尿症状较模型组有所改善,大鼠活动增加。与正常对照组比较,模型组大鼠体重降低,白细胞计数(WBC)、红细胞(RBC)、血小板(PLT)、天冬氨酸转氨酶(AST)、尿素氮(BUN)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB),糖负荷后30、60、90、120 min大鼠血糖,胰岛素抵抗指数(HOMA-IR)、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、左室舒张末期内径(LVEDD)、左室收缩末期内径(LVESD)、HMGB1 mRNA、NF-κB p65 mRNA均升高,高密度脂蛋白胆固醇(HDL-C)、缩短分数(FS)、射血分数(EF)均降低,差异有统计学意义(P<0.05),且模型组大鼠在糖负荷后60 min血糖达到峰值。与模型组比较,阳性对照组、黑醋栗组大鼠体重降低,WBC、RBC、PLT、AST、BUN、CK、CK-MB、血糖、HOMA-IR、TC、TG、LDL-C、LVEDD、LVESD均降低,HDL-C、FS、EF均升高,差异有统计学意义(P<0.05)。结论 黑醋栗可改善T2DM大鼠糖脂代谢水平、心功能和心肌肥厚,这可能是通过抑制炎症通路HMGB1/NF-κB来实现的。
Objective To explore the possible mechanism by which blackcurrant improves cardiac function and myocardial hypertrophy in type 2 diabetic(T2DM)rats.Methods Forty adult male Sprague-Dawley(SD)rats were randomly divided into 4 groups:normal control group,model group(T2DM model),positive control group(T2DM model+liraglutide),and blackcurrant group(T2DM model+blackcurrant intervention).T2DM was induced by feeding rats with high-fat and high-glucose diet.At 10 weeks after model establishment,rats were observed for behavioral changes,their body weights were determined and oral glucose tolerance tests(OGTTs)were performed.The cardiac function of rats was detected by ultrasound.A full-automatic biochemical analyzer was utilized to test blood routine indices,lipid index,liver and kidney biochemical indices,and myocardial enzyme indexes.The mRNA of high mobility group box protein 1(HMGB1)and nuclear factor kappa B(NF-κB)in rat myocardial tissue were detected by real-time reverse transcriptase polymerase chain reaction(RT-PCR).Results Behavioral observation denoted that rats in normal control group developed normally with gloss hair and being active to external stimulation,and rats in model group suffered from dry hair,symptoms of polydrinking,polyphagia and polyuria,and decreased activity,whereas the above mentioned symptoms were all alleviated in positive control group and blackcurrant group,and rats in two groups had increased activity.Compared with normal control group,rats in model groups had a decrease in body mass index,and an increase in white blood cell count(WBC),red blood cell count(RBC),platelet(PLT),aspartate aminotransferase(AST),blood urea nitrogen(BUN),creatine kinase(CK),creatine kinase-MB isoenzyme(CK-MB),blood glucose at 30,60,90,and 120 min after oral glucose loading,
