摘要
以100份豌豆种质为材料,采用SDS-PAGE电泳技术获取种子盐溶蛋白和水溶蛋白图谱,建立数据矩阵并进行遗传多态性、品种聚类分析。结果表明,两类蛋白均分离出36条条带,其中多态性条带30条。盐溶蛋白多态性条带平均显带次数为60.70、变异系数为48.14%,供试品种平均显带数为18.21、变异系数为15.14%;水溶蛋白多态性条带平均显带次数为63.43、变异系数为39.22%,供试品种平均显带数为19.03、变异系数为15.65%。电泳条带数目及分布差异使品种间带型变异显著,表明豌豆种子贮藏蛋白具有丰富遗传多态性,可作为有效的遗传标记。基于60条多态性条带信息进行UPGMA聚类分析,在遗传相似系数0.57处将100份豌豆划分为7个类群,81个品种聚于品种群Ⅰ,19个遗传差异大的品种分散于其他群,主成分分析二维散点图进一步印证了品种分布特征。
Taking 100 Pisum sativum germplasms as materials,the salt-soluble protein and water-soluble protein maps of seeds were obtained by SDS-PAGE electrophoresis technology.The data matrix was established,genetic polymorphisms were analyzed,and variety clustering was carried out.The results showed that 36 bands were isolated from both types of proteins,among which 30 were polymorphic bands.The average number of bands of the salt-soluble protein polymorphism was 60.70 and the coefficient of variation was 48.14%,while the average number of bands of the tested varieties was 18.21 and the coefficient of variation was 15.14%.The average number of bandings of water-soluble protein polymorphism bands was 63.43,with a coefficient of variation of 39.22%,while the average number of bandings of the tested varieties was 19.03,with a coefficient of variation of 15.65%.The differences in the number and distribution of electrophoretic bands led to significant band type variations among varieties,indicating that Pisum sativum seed storage proteins had rich genetic polymorphisms and could be used as effective genetic markers.Based on the information of 60 polymorphic bands,UPGMA cluster analysis was conducted.At a genetic similarity coefficient of 0.57,100 Pisum sativum samples were divided into seven groups.81 varieties were clustered in variety GroupⅠ,while 19 varieties with significant genetic differences were dispersed in other groups.The two-dimensional plot of principal component analysis further confirmed the distribution characteristics of the varieties.
