释迦果多糖水提工艺优化及体外经皮吸收特性研究

Optimization of the aqueous extraction process and in vitro transdermal absorption of polysaccharides from Sakya fruit

摘要 以释迦果果肉为原料,多糖得率为响应值,利用超声辅助热水浸提法进行提取,考察4个因素,并结合正交试验优化释迦果多糖水提液的提取工艺,利用离子色谱技术分析其单糖组成,通过DPPH、·OH、FRAP实验评价其抗氧化活性。采用Franz扩散池进行离体经皮实验,测定其累计渗透量,计算渗透动力学参数。结果显示,在最佳提取条件下,多糖得率为50.57%±0.02%。离子色谱分析表明,多糖主要成分为阿拉伯糖(0.6%)、葡萄糖(40.5%)和果糖(58.9%)。抗氧化性能评价结果表明,当添加80μL提取液时,DPPH自由基清除率为94.13%±0.81%;加入600μL时,羟自由基清除率为93.33%±0.92%,FRAP值为(38.07±1.30)mmol/L。经皮吸收实验结果显示,释迦果多糖水提液在75%和100%浓度下的皮肤滞留率分别为4.65%±1.52%和12.37%±2.14%,12 h累积渗透率分别为57.43%±1.08%和74.24%±0.74%。经皮吸收的动力学分析表明,该过程符合零级动力学特征,即在稳态条件下药物以恒定速率释放,符合Fick扩散定律。 The pulp of Sakya fruit was used as raw material,and the polysaccharide yield was the response value,and the extraction was carried out by using ultrasound-assisted hot water immersion method,four factors were investigated and combined with the orthogonal test to optimize the process of aqueous extraction of polysaccharides from Sakya fruit,and the composition of the monosaccharides was analyzed by using ion chromatography,and the antioxidant activity was evaluated by the DPPH,·OH,and FRAP assays.The Franz diffusion cell was used for the ex vivo transdermal assay to determine the cumulative permeation volume and calculate the permeation kinetic parameters.The results showed that the polysaccharide yield was 50.57%±0.02% under the optimal extraction conditions.Ion chromatographic analysis showed that the main components of polysaccharides were arabinose(0.6%),glucose(40.5%)and fructose(58.9%).Evaluation of antioxidant properties showed that the DPPH radical scavenging rate was 94.13%±0.81% when 80μL of the extract was added,and the hydroxyl radical scavenging rate was 93.33%±0.92% when 600μL was added,with a FRAP value of(38.07±1.30)mmol/L.Results of the transdermal absorption experiments showed that,at the concentrations of 75%and 100%,the Sakya Fruit polysaccharide aqueous extract had a skin retention of 4.65%±1.52% and 12.37%±2.14% at 75% and 100% concentrations,respectively,and a 12 h cumulative permeation rate of 57.43%±1.08% and 74.24%±0.74%,respectively.The kinetic analysis of percutaneous absorption showed that the process conformed to the zero-grade kinetic characteristics,i.e.,the drug was released at a constant rate under steady state conditions in accordance with Fick’s law of diffusion.
