摘要
为满足清醒小鼠的脑成像需求,研发了一种小鼠脑固定装置,并基于该装置建立了一套双光子显微镜活体小鼠脑样本制备方法。该方法涵盖小鼠麻醉和固定、颅骨开窗、头部固定器安装等关键步骤。利用此方法制备的小鼠脑样本,在双光子显微镜下观测脑内钙信号、血管网络及神经元结构时,可获得高信噪比的清晰图像。该活体小鼠脑样本制备方法为深入探究生理状态下鼠脑结构与功能提供了有力支撑。
To meet the imaging needs of awake mice,a novel mouse brain fixation device was independently developed.A detailed method for brain sample preparation and optimization was established based on this device,which is suitable for two-photon microscopic brain imaging experiments in awake mice.When we study signals such as intracranial calcium signals,vascular networks,and neuronal structures,we are possible to obtain clear images with a high signal-to-noise ratio.The method for preparing brain samples from living mice provides strong support for in-depth exploration of the structure and function of the mouse brain under physiological conditions.
