摘要
该文建立小鼠辐射损伤模型,对小鼠体质量生长曲线、外周全血白细胞数、脏器指数、白细胞介素-6(interleukin-6,IL-6)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)含量等指标进行分析,同时对免疫器官进行苏木精-伊红(hematoxylin-eosin,HE)染色病理分析,检测脾脏组织Toll样受体4(Toll-like receptor 4,TLR4)、核因子κB(nuclear factor kappa-B,NF-κB)P65的表达,研究赤松茸多糖缓解小鼠辐射损伤的作用机制。结果表明,赤松茸多糖不仅可以有效提高外周全血白细胞数和脏器指数,还能有效抑制炎症因子分泌,减轻脾脏、胸腺、肝脏和结肠组织的损伤,降低脾脏组织TLR4、NF-κB P65的mRNA表达量和蛋白表达。综上,赤松茸多糖可有效改善辐射对小鼠脏器所致损伤,通过TLR4/NF-κB通路抑制辐射所致炎症反应,是一种具有开发潜力的辐射损伤防护活性物质。
A mouse model of radiation damage was established.The body weight growth curve,white blood cell count,organ indexes,and levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)of mice were analyzed.The immune organs were stained with hematoxylineosin(HE).The expression levels of Tolllike receptor 4(TLR4)and nuclear factor kappaB(NF-κB)P65 in the spleen tissue were determined to study the mechanism of Stropharia rugosoannulata polysaccharide in alleviating radiation damage in mice.The results showed that S.rugosoannulata polysaccharide not only effectively increased the peripheral white blood cell count and organ indexes but also effectively inhibited the secretion of inflammatory factors,reduced the damage of the spleen,thymus,liver,and colon tissue,and down-regulated the mRNA and protein levels of TLR4 and NF-κB P65 in the spleen tissue.In summary,S.rugosoannulata polysaccharide effectively alleviated the organ damage caused by radiation and inhibited the inflammatory responses caused by radiation by acting on the TLR4/NF-κB pathway in mice.It demonstrated potential as a protective agent for radiation damage.
