高效液相色谱法同时测定荷叶降脂煎煎煮液及其主要原料药中7种成分

Simultaneous Determination of 7 Substances in Lotus Leaf Lipid Reduction Decoction and its Main Original Herbs by High Performance Liquid Chromatography

摘要 目的:建立荷叶降脂煎煎煮液及其主要原料药中7种成分大黄酚、橙黄决明素、荷叶碱、熊果酸、23-乙酰泽泻醇B、丹参酮IIA、苍术素的液相色谱分析方法。方法:使用Supersil ODS-2色谱柱,以水-乙腈为流动相梯度洗脱,检测波长220、260、280、340 nm。样品用甲醇超声提取,提取液过滤膜后进行分析。结果:7种成分得到良好分离,在线性范围内具有良好的线性关系(R2>0.998),检测限和定量限分别为0.002~0.176μg/mL和0.007~0.588μg/mL。加标回收率在80.1%~113.9%,RSD值均小于3%,并成功用于实际样品分析。结论:该方法具有简洁、准确和高效等特点,可为荷叶降脂煎制剂质量标准研究提供参考。 Objective:Development of a novel HPLC method for simultaneous determination of 7 substances nuciferine,chtysophanol,aurantion-obtusin,ursolic acid,alisol B 23-monoacetate,tanshinone IIA,atractylodin in Lotus Leaf Lipid Reduction Decoction and its main original herbs.Methods:The separation column was Supersil ODS-2,and mobile phase was water-acetonitrile with gradient elution.The detection wavelengths were 220,260,280,340 nm.The sample was extracted by methanol before analysis.Results:The 7 substances were well separated with good linearity(R2>0.998)in the linear ranges.And the limits of detection(LOD)and limits of quantification(LOQ)were in the ranges of 0.002-0.176μg/mL and 0.007-0.588μg/mL,respectively.The recoveries are in the range of 80.1%-113.9%,with RSD<3%.The method was also successfully applied to real sample analysis.Conclusion:This method is concise,accurate and efficient,which can provide a reference for the study of the quality control of the Lotus Leaf Lipid Reduction Decoction.
