摘要
目的建立茯苓四种药用部位茯神、茯苓、赤茯苓、茯苓皮中26种无机元素的电感耦合等离子体质谱(ICP-MS)分析方法,并对其无机元素分布特征进行比较.方法以硝酸为消解试剂,微波消解法处理样品;以Sc、Ge、Rh、In、Tb、Bi元素为内标,内标标准曲线法进行样品测定.根据无机元素测定结果,采用直观分析、聚类分析(HCA)和主成分分析(PCA)和正交偏最小二乘法判别分析(OPLS-DA)对茯苓四种药用部位共31批样品进行比较.结果26种无机元素的浓度与比率在测定浓度范围内线性关系良好,方法重复性的RSD均小于6.4%、回收率均在94.0%~115.9%内.茯苓皮中26种无机元素总量最大,且与其他三种部位差别最大,主要差别元素是Al、V、Fe、Se、Ca、Ba、Co、Rb、Cs、Mg、As、Cu;茯神和茯苓无机元素含量分布接近;Na、Mg、Al、P、K、Ca、Fe元素在各部位药材中的含量高于其他元素.结论本研究建立的方法可用于茯苓不同药用部位中无机元素的测定和比较,为茯苓四种药用部位的质量评价和功效差异的阐明奠定了基础.
Objective To establish an inductively coupled plasma mass spectrometry(ICP-MS)method for the analysis of 26 inorganic elements in Poria cum Radix Pini,White Poria,Rubra Poria and Poriae Cutis,and to compare the distribution characteristics of inorganic elements.Methods The samples were treated by microwave digestion with nitric acid as digestion reagent and were determined by internal standard curve method with Sc,Ge,Rh,In,Tb and Bi as internal standards.According to the determination results of inorganic elements,31 batches of samples from four medicinal parts of Poria cocos(Schw.)Wolf were compared by intuitive analysis,cluster analysis(HCA),principal component analysis(PCA)and orthogonal partial least square discriminant analysis(OPLS-DA).Results The concentration and ratio of 26 inorganic elements had a good linear relationship in the concentration range.The repeatability RSD of the method was less than 6.4%,and the recovery was in the range of 94.0%-115.9%.The total amount of 26 inorganic elements in Poriae Cutis was the largest,and it was the most different from the other three parts.The main difference elements were Al,V,Fe,Se,Ca,Ba,Co,Rb,Cs,Mg,As and Cu;the content distribution of inorganic elements in Poria cum Radix Pini and White Poria was similar;the contents of Na,Mg,Al,P,K,Ca and Fe in different parts of medicinal materials were higher than those of other elements.Conclusion The established method can be used for the determination and comparison of inorganic elements in different medicinal parts of Poria cocos(Schw.)Wolf,which lays a foundation for the quality evaluation and efficacy difference of four medicinal parts of Poria cocos(Schw.)Wolf.
