摘要
目的乌天麻为高海拔产名贵中药材,目前缺乏温度和种子成熟度对其种子萌发影响的研究。阐明这两种因素对种子萌发的影响,对乌天麻有性繁殖防止天麻退化有重要理论和实际应用价值。方法本实验对以乌天麻为主的3种天麻变型进行2,3,5-三苯基氯化四氮唑(TTC)染色和低温共生萌发实验,测定不同成熟度种子的性状和萌发指标,采用主成分分析和隶属函数值法对种子活力指标进行综合评价分析,并采用冗余分析(RDA)揭示种子萌发和温度及种子成熟度的相关性。结果乌天麻和绿天麻种子细长型,红天麻短粗型,且乌天麻种胚体积最大;不同天麻的低温共生萌发结果存在差异(P<0.05)。结论温度显著影响种子萌发,而成熟度影响相对较小,结合综合评价值,明确了乌天麻最优萌发组合为16℃-将裂果,为乌天麻规模化生产提供了科学依据。
OBJECTIVE To elucidate the effects of temperature and seed maturity on seed germination of Gastrodia elata Bl.f.glauca and provide theoretical and practical value for the sexual reproduction of G.elata Bl.f.glauca and the prevention of its degen-eration.METHODS 2,3,5-triphenyltetrazolium chloride(TTC)staining and low-temperature symbiotic germination experiments were performed on three formae of G.elata(G.elata Bl.f.glauca as the primary focus).Seed traits and germination indices were quantified across maturity stages.A combined approach of Principal Component Analysis(PCA)and membership function valuation was applied for comprehensive seed vigor assessment.Furthermore,Redundancy Analysis(RDA)was utilized to reveal the correla-tions among seed germination,temperature,and seed maturity.RESULTS The seeds of G.elata Bl.f.glauca and G.elata Bl.f.viridis exhibited an elongated morphology in contrast to the shorter and thicker seeds of G.elata Bl.f.elata,and G.elata Bl.f.glau-ca possessed the largest embryo volume among the variants.The low-temperature symbiotic germination outcomes varied significantly a-mong the different G.elata variants(P<0.05).CONCLUSION Temperature significantly affects seed germination,whereas the impact of maturity is relatively minor.Based on the comprehensive evaluation values,the optimal germination combination for G.elata Bl.f.glauca is determined to be 16℃-near-dehiscent fruit,providing a scientific basis for the large-scale production of G.elata Bl.f.glauca.
