含“益宫清”奶牛血清缓解奶牛子宫内膜上皮细胞炎症损伤的作用机制

摘要 选用大庆市杜尔伯特牛场奶牛,筛选“益宫清”治疗奶牛子宫内膜炎的最佳剂量,并制备含药血清。将含药血清作用于经LPS处理的奶牛子宫内膜上皮细胞(BEND),通过蛋白组学分析筛选差异蛋白,探究其作用机制。结果表明:高剂量组(质量浓度为220 g·L^(-1))治疗效果最佳,并按此剂量制备含药血清;体外试验表明,益宫清可显著降低LPS诱导的BEND细胞炎症因子表达水平;蛋白组学分析结果共鉴定出7 342个相关差异蛋白;GO富集分析显示,差异蛋白主要富集在生物进程和抗氧化过程等;KEGG聚类分析显示,差异蛋白主要涉及PPAR、AMPK、NFκB等信号通路;验证差异显著蛋白NFκB2、NκIRAS2,与蛋白质组学结果一致,表明益宫清可能通过上调NFκB2及下调NκIRAS2蛋白发挥抗炎作用。 Selecting dairy cows from Duerbote's farm cattle in Daqing City,screening the optimal dosage of"Yigongqing"for treating bovine endometritis,and preparing a drug containing serum.The drug-containing serum was applied to LPS-treated bovine endometrial epithelial cells(BEND),and by analyzing proteomics and screening differential proteins,explored their mechanisms of action.The results showed that the high-dose group was the optimal dosage for treatment,and drug-containing serum was prepared at this dosage.In vitro experiments demonstrated that"Yigongqing"could reduce the contents of inflammatory cytokines induced by LPS in BEND cells.Proteomic analysis identified 7342 differentially expressed proteins;GO enrichment analysis revealed that the differentially expressed proteins were primarily enriched in biological processes and antioxidant processes;KEGG analysis showed that these proteins were mainly involved in PPAR,AMPK,and NFκB signaling pathways;the validation of the significantly different proteins NFκB2 and NκIRAS2 was consistent with the proteomic results.This indicated that"Yigongqing"might exert anti-inflammatory effects by upregulating NFκB2 and downregulating NκIRAS2 protein.
