局部脉搏波速度对动脉壁弹性评估的大鼠实验研究

摘要 目的探讨用超声设备测量局部脉搏波速度(PWV)评估动脉壁弹性的价值。方法选取10周龄自发性高血压大鼠(SHR)及10周龄正常血压Wistar-Kyoto(WKY)大鼠各8只为研究对象。测量大鼠血压,利用超声诊断仪检测大鼠腹主动脉局部弹性。数据采集完成后解剖大鼠,截取一段腹主动脉,进行病理学染色,判断腹主动脉管壁弹性情况,并根据病理组织学结果分组,分为动脉壁正常组及动脉壁硬化组。采用非参数Mann-Whitney U检验比较两组大鼠的血管弹性指标、收缩压、舒张压的差异。结果去除数据采集不完整的大鼠,实验最终纳入12只大鼠,WKY大鼠5只,SHR大鼠7只。病理组织学结果显示动脉壁正常组6例,动脉壁硬化组6例。动脉壁硬化组大鼠的PWV和Ep分别为(6.14±1.94)m/s、(91.54±63.07)Kp,均明显高于动脉壁正常组[(3.59±0.46)m/s、(35.57±13.26)Kp],动脉壁硬化组CC和DC分别为(0.06±0.03)mm^(2)/pa、(30.03±12.79)1/mpa,均明显低于动脉壁正常组[(0.15±0.05)mm^(2)/pa、(68.50±28.08)1/mpa],差异均有统计学意义(P<0.05);两组大鼠的收缩压、舒张压比较,差异均无统计学意义(U=20.000、21.000,P=0.750、0.630)。结论大鼠实验表明,超声测量局部PWV能准确评估动脉壁弹性,早期发现动脉硬化,临床上可以用此方法对心脑血管高危人群进行动脉壁弹性筛查,早期发现血管病变。 Objective To investigate the value of local pulse wave velocity(PWV)measurement by ultrasonic equipment in assessing arterial wall elasticity.Methods Spontaneously hypertensive rats(SHR)at 10 weeks of age and Wistar-Kyoto rats(WKY)at 10 weeks of age were selected as the study subjects.The number of subjects was 8 each.The blood pressure of the rats was measured,and the local elasticity of the abdominal aorta of the rats was detected by ultrasonic equipment.After data collection,the rats were dissected,and a section of abdominal aorta was extracted for pathological staining to determine the elasticity of abdominal aorta wall.According to the histopathological results,the rats were divided into the normal arterial wall group and the arteriosclerosis group.Non-parametric Mann-Whitney U test was used to compare the differences in vascular elasticity indexes,systolic blood pressure and diastolic blood pressure between the two groups.Results The incomplete data of rats were removed,12 rats were included in the study,including 5 WKY rats and 7 SHR rats.The histopathological results showed that the aortic wall was normal in 6 rats and the arterial wall was sclerotic in 6 rats.The PWV and Ep in the arteriosclerosis group were(6.14±1.94)m/s and(91.54±63.07)Kp,respectively,which were significantly higher than those in the normal arterial wall group[(3.59±0.46)m/s and(35.57±13.26)Kp],the CC and DC in the arteriosclerosis group were(0.06±0.03)mm^(2)/pa and(30.03±12.79)1/mpa,respectively,which were significantly lower than those in the normal arterial wall group[(0.15±0.05)mm^(2)/pa and(68.50±28.08)1/mpa],the differences were statistically significant(P<0.05).While the differences in systolic and diastolic blood pressure were not statistically significant between the two groups of rats(U=20.000,21.000;P=0.750,0.630).Conclusion Experiments in rats show that measuring local PWV with ultrasonic equipment can accurately assess the early lesions of the artery wall.Clinically,this method can be used to screen the artery wall elasticity of high-risk groups of cardiovascular and cerebrovascular diseases and to detect the vascular lesions early.
