摘要
间充质干细胞(MSCs)凭借多向分化潜能,成为细胞治疗与组织工程领域至关重要的种子细胞。线粒体自噬在干细胞功能调控中扮演着核心角色。适度的线粒体自噬可促进线粒体更新代谢,维持MSCs的干性与增殖能力。同时,它还参与调控MSCs的代谢与免疫过程,有助于维持细胞内环境稳态,延缓细胞衰老,这对于口腔组织修复过程中MSCs持续发挥作用意义重大。目前,学界公认线粒体自噬调控干细胞功能的机制包括泛素依赖途径、非泛素依赖途径和非经典途径。本综述对线粒体自噬在间充质干细胞功能调控方面的作用及机制进行了全面梳理,详细阐述了线粒体自噬的定义、功能以及其调控间充质干细胞功能的具体过程与相关机制。鉴于线粒体自噬在MSCs功能调控中的重要性,未来研究应着重聚焦于调控机制方面,如溶酶体驱动的降解途径等。在口腔医学领域,这可能为解决牙周炎导致的牙槽骨吸收、口腔黏膜疾病的修复等问题提供新的思路。同时,结合线粒体自噬在神经退行性疾病、心血管疾病等领域展现出的潜在治疗价值,有望为基于MSCs的再生医学和细胞治疗发掘新的干预靶点,推动口腔组织修复和再生治疗的发展。
Mesenchymal stem cells(MSCs),due to their multipotent differentiation potential,have become critical seed cells in the fields of cell therapy and tissue engineering.Mitophagy plays an important role in maintaining cell homeostasis especially in MSCs.Moderate mitophagy facilitates mitochondrial renewal and metabolism,thus maintaining the stemness and proliferation ability of MSCs.Additionally,mitophagy participate in the regulation of metabolism and immune responses of MSCs,contributing to intracellular homeostasis and delaying cellular senescence,which plays an highly significant role in the process of oral tissue repair.The underlying regulatory mechanisms include ubiquitin-dependent pathways,non-ubiquitindependent pathways and non-classical pathways.This review provides a comprehensive summary of the roles and mechanisms of mitophagy in regulating MSCs functions.It elaborates on the definition and function of mitophagy,as well as the specific processes and associated mechanisms through which it regulates MSCs behavior.Considering the importance of mitophagy in modulating the function of MSCs,this review also proposes that future research focus on the regulatory mechanism(such as lysosome-driven degradation pathways).In the field of stomatology,such investigations may offer novel insights and innovative solutions for addressing the challenges of alveolar bone loss caused by periodontitis and the repair of oral mucosal diseases.Meanwhile,combined with potential therapeutic value in neurodegenerative diseases,cardiovascular diseases,and other fields,it is expected to find new insights and therapeutic targets for MSC-based regenerative medicine and cell therapy,and promote the development of oral tissue repair and regenerative therapy.
