摘要
目的:探讨ABw07亚型血清学和分子生物学特点。方法:血清学用试管法和微柱凝胶卡式法检测血型,分子生物学用荧光PCR法进行ABO基因分型,后用PCR扩增ABO基因外显子后进行直接测序,并对先证者进行家系调查。运用Alphafold3.0软件建立3D模型,预测基因突变导致蛋白结构的影响。结果:血清学先证者红细胞与抗-B反应呈弱凝集,反定型为AB型;基因分型结果显示:先证者及父亲为ABw07,直接测序结果与A1.01/B.01相比发现特异性位点c.467C>T、c.1055G>A发生碱基突变。结论:在山东济南地区人群中检测到Bw07的等位基因。血清学结合分子生物学方法能够正确的鉴定血型,血型的准确性是保障临床用血安全的前提。
Objective To investigate the serological and molecular biological characteristics of the ABw07 subtype.Methods Blood group was detected by the tube tube method and microcolumn gel cassette method in serology.ABO genotyping was performed by fluorescent PCR method in molecular biology.ABO exons were amplified by PCR and sequenced directly.Alphafold software was used to build 3D models to predict the impact of gene mutation on protein structure.Results The erythrocytes of the progenitor showed weak agglutination with the anti-B reaction,and the reverse pattern was AB type.Genotyping results The progentor and father were ABw07,and the direct sequencing results compared with A1.01/B.01 showed that specific sites c.467C>T and c.1055G>A had base mutations.Conclusion The allele of Bw07 was detected for the first time in Jinan,Shandong Province.The method of serology combined with molecular biology can identify blood type correctly,and the accuracy of blood type is the premise of ensuring the safety of blood for clinical use.
