摘要
目的 基于Janus激酶1(JAK1)/信号转导和转录活化因子6(STAT6)信号通路观察捏脊疗法对支气管哮喘幼鼠气道炎症及黏液分泌的影响。方法 选取BALB/c雌性幼鼠40只,按体重随机分为正常组、模型组、地塞米松组、捏脊组,每组10只。除正常组外,其余组幼鼠采用卵清蛋白(OVA)腹腔注射致敏+雾化激发法建立支气管哮喘模型。造模成功后,地塞米松组给予地塞米松1 mg/kg腹腔注射,捏脊组给予捏脊法干预,均1次/d,连续干预4周。无创肺功能检测系统测定幼鼠增强呼吸间歇(Penh)值,ELISA法测定血清中白细胞介素-4(IL-4)、白细胞介素-13(IL-13)、肿瘤坏死因子-α(TNF-α)水平和血浆中免疫球蛋白E(IgE)水平,计数肺泡灌洗液中炎症细胞总数,计算肺泡灌洗液中嗜酸性粒细胞比例,ELISA法测定肺泡灌洗液中黏蛋白5AC(MUC5AC)、MUC5B、细胞间黏附分子-1(ICAM-1)水平,Western blot法及RT-PCR法检测肺组织中JAK1、STAT6、细胞因子信号传导抑制蛋白3(SOCS3)、SOCS5蛋白和mRNA表达情况。结果与正常组比较,模型组幼鼠吸入不同浓度乙酰甲胆碱溶液后Penh,血清IL-4、IL-13、TNF-α水平,血浆IgE水平,肺泡灌洗液中炎症细胞总数、嗜酸性粒细胞比例及MUC5AC、MUC5B、ICAM-1水平,肺组织中JAK1、STAT6、SOCS3蛋白及mRNA相对表达量均明显升高(P均<0.05),肺组织中SOCS5蛋白及mRNA相对表达量均明显降低(P均<0.05);与模型组比较,地塞米松组和捏脊组幼鼠吸入不同浓度乙酰甲胆碱溶液后Penh,血清IL-4、IL-13、TNF-α水平,血浆IgE水平,肺泡灌洗液中炎症细胞总数、嗜酸性粒细胞比例及MUC5AC、MUC5B、ICAM-1水平,肺组织中JAK1、STAT6、SOCS3蛋白及mRNA相对表达量均明显降低(P均<0.05),肺组织中SOCS5蛋白及mRNA相对表达量均明显升高(P均<0.05)。结论 捏脊疗法可明显抑制支气管哮喘幼鼠气道炎症反应,改善黏液高分泌状态,其机制可能与抑制JAK1/STAT6信号通路活化相关。
Objective It is to observe the effect of chiropractic therapy on airway inflammation and mucus secretion in young mice with bronchial asthma via Janus kinase 1(JAK1)/signal transducer and activator of transcription 6(STAT6)signaling pathway.Methods Forty female BALB/c young rats were selected and randomly divided into normal group,model group,dexamethasone group and chiropractic therapy group according to their weight,with 10 mice in each group.The mice of all groups except for the normal group were sensitized by intraperitoneal injection of ovalbumin(OVA)and challenged via nebulization to establish models of bronchial asthma.After successful modeling,the dexamethasone group was given dexamethasone(1 mg/kg)by intraperitoneal injection,the chiropractic therapy group was treated with chiropractic therapy group,all once daily,continuously treated for 4 weeks.The enhanced pause(Penh value)of the young mice was detected by non-invasive pulmonary function detection system,the serum levels of interleukin-4(IL-4),interleukin-13(IL-13)and tumor necrosis factor-α(TNF-α),as well as plasma levels of immunoglobulin E(IgE)were measured by ELISA,the total number of inflammatory cells in bronchoalveolar lavage fluid(BALF)was counted,and the proportion of eosinophils was calculated,the levels of mucin 5AC(MUC5AC),MUC5B,and intercellular adhesion molecule-1(ICAM-1)in BALF were measured by ELISA,The protein and mRNA expressions of JAK1,STAT6,suppressor of cytokine signaling 3(SOCS3)and SOCS5 in lung tissue were detected by Western blot and RT-PCR,respectively.Results Compared with the normal group,after inhaling different concentrations of acetylcholine solution,the Penh value,serum levels of IL-4,IL-13 and TNF-α,plasma level of IgE,total inflammatory cell count,eosinophil proportion and levels of MUC5AC,MUC5B,ICAM-1 in BALF,as well as the relative protein and mRNA expressions of JAK1,STAT6 and SOCS3 in lung tissue of young mice in the model group were significantly increased(all P<0.05),while the relative protein and mRNA expressions of SOCS5 in lung tissue were significantly decreased(both P<0.05).Compared with the model group,after inhaling different concentrations of acetylcholine solution,the Penh value,serum levels of IL-4,IL-13 and TNF-α,plasma level of IgE,total inflammatory cell count,eosinophil proportion and levels of MUC5AC,MUC5B,ICAM-1 in BALF,as well as the relative protein and mRNA expressions of JAK1,STAT6 and SOCS3 in lung tissue of young mice in the dexamethasone group and chiropractic therapy group were significantly decreased(all P<0.05),while the relative protein and mRNA expressions of SOCS5 in lung tissue were significantly increased(both P<0.05).Conclusion Chiropractic therapy can significantly inhibit airway inflammation and ameliorate mucus hypersecretion in young mice with bronchial asthma.The mechanism may be associated with the inhibition of JAK1/STAT6 signaling pathway activation.
