隐丹参酮调节TXNIP/NLRP3信号通路对肺炎克雷伯菌肺炎大鼠炎症反应的影响

The effect of cryptotanshinone on the inflammatory response of Klebsiella pneumoniae pneumonia rats by regulating theTXNIP/NLRP3 signaling pathway

摘要 目的 探究隐丹参酮(Cryp)调节TXNIP/NLRP3信号通路对肺炎克雷伯菌肺炎大鼠炎症反应的影响。方法 将大鼠随机分为NC组、Model组、Cryp-L组、Cryp-H组、Cryp-H+氧化三甲胺(TMAO)(NLRP3激活剂)组和左氧氟沙星组。血气分析仪检测PaO_(2)和PaCO_(2);计算肺组织湿/干(W/D)比;观察肺组织形态;检测肺组织细胞凋亡、IL-1β、IL-6、IL-18、TNF-α水平、MDA含量、MPO和SOD活性、SP-A、SP-D、TXNIP、NLRP3和caspase-1蛋白表达水平。结果 与NC组比较,Model组大鼠肺组织有明显损伤,PaO_(2)和OI水平、SOD活性、SP-A和SP-D蛋白表达水平降低,PaCO_(2)水平、肺组织病理评分、IL-1β、IL-6、IL-18和TNF-α水平、W/D值、MDA含量和MPO活性、TXNIP、NLRP3和caspase-1蛋白表达水平升高(P<0.05);与Model组比较,Cryp-L组、Cryp-H组和左氧氟沙星组大鼠肺组织损伤程度减轻,PaO_(2)和OI水平、SOD活性、SP-A和SP-D蛋白表达水平升高,PaCO_(2)水平、肺组织病理评分、IL-1β、IL-6、IL-18和TNF-α水平、W/D值、MDA含量和MPO活性、TXNIP、NLRP3和caspase-1蛋白表达水平降低(P<0.05);TMAO可降低Cryp对Kp肺炎大鼠的改善作用(P<0.05);Cryp-H组和左氧氟沙星组上述各指标处于同一水平(P>0.05)。结论 Cryp可降低Kp肺炎大鼠炎症和肺损伤,可能与抑制TXNIP/NLRP3信号通路有关。 Objective To discuss the effect of cryptotanshinone(Cryp)on the inflammatory response of Klebsiella pneumoniae pneumonia rats by regulating the TXNIP/NLRP3 signaling pathway.MethodsRats werestochastically separated into NC group,Model group,Cryp-L group,Cryp-H group,Cryp-H+trimetlylamineoxide(TMAO)(NLRP3 activator)group,and levofloxacin group.Blood gas analyzer was used to detect PaO_(2) and PaCO_(2).The wet/dry(W/D)ratio of lung tissue was calculated.The morphology of lung tissue as observed.The apoptosis of lung tissue cells,the Il-1p,IL-6,IL-18,TNF-αlevels,MDA content,MPO and SOD activities,SP-A,SP-D,TXNIP,NLRP3,and caspase-1 proteins were measured.Results Compared with the NC group,the Model group showed prominent injury to rat lung tissue.The PaO,and OI levels,SOD activity,SP-A and SP-D proteins decreased,while the PaCO_(2) level,lung tissue pathological score,IL-1p,IL-6,IL-18 and TNF-αlevels,W/D value,MDA content,MPO activity,TXNIP,NLRP3 and caspase-l proteins increased(P<0.05).Compared with the Model group,the Cryp-L group,Cryp-H group,and levofloxacin group showed a reduction in the degree of lung tissue injury in rats.The PaO_(2) and OI levels,SOD activity,SP-A and SP-D proteins increased,while the PaCO_(2) level,lung tissue pathological score,IL-1p,IL-6,IL-18 and TNF-αlevels,W/D value,MDA content,MPO activity,TXNIP,NLRP3 and caspase-1 proteins decreased(P<0.05).TMAO could reduce the improvement effect of Cryp on Kp pneumonia rats(P<0.05).The above indicators of Cryp-H group and levofloxacin group were at the same level(P>0.05).Conclusion Cryp can reduce inflammation and lung injury in Kp pneumonia rats,which may be related to the inhibition of TXNIP/NLRP3 signaling pathway.
