摘要
【目的】通过对凉山黑绵羊和湖羊全基因组重测序,解析凉山黑绵羊毛色形成的主效基因,为凉山黑绵羊种质资源评价和开发利用提供理论基础。【方法】19个凉山黑绵羊和28个湖羊的DNA样本经BGISEQ DNBSEQ-T7platform平台进行测序,整合NCBI数据库中的巴美肉羊和无角陶赛特羊的测序数据进行PCA、选择信号和KEGG等分析。【结果】PCA结果显示凉山黑绵羊独立聚类,通过F_(ST)和θπ分析在凉山黑绵羊与湖羊、巴美肉羊、无角陶赛特羊中筛选出了U6、JAZF1和SLC26A1等受选择基因。基因型可视化分析揭示,凉山黑绵羊的KIT基因野生型频率显著高于白色绵羊品种;另外,凉山黑绵羊的TRPM1、TMEM18、MC1R和ASIP基因的纯合突变频率均显著高于白色绵羊品种。【结论】KIT、MC1R、ASIP、TMEM18和TRPM1基因是凉山黑绵羊毛色形成的关键基因,研究结果为深入理解其毛色遗传分子机制提供理论依据。
【Objective】This study aimed to analyze the main effector genes associated with wool color formation in Liangshan Black Sheep by the whole genome resequencing of Liangshan Black Sheep and Hu Sheep,which provided a theoretical foundation for evaluating and utilizing the genetic resources of Liangshan Black Sheep.【Method】DNA samples from 19 Liangshan Black Sheep and 28 Hu Sheep were sequenced using the BGISEQ DNBSEQ-T7 platform.These data were integrated with resequencing data from Bamei Sheep and Poll Dorset sourced from the NCBI database to conduct principal component analysis(PCA),selection signal analysis and KEGG analysis.【Result】PCA revealed a distinct separation of Liangshan Black Sheep from three white sheep breeds,resulting in the formation of independent clusters.By F_(ST) andθπanalysis,the selected genes,including U6,JAZF1,and SLC26A1,were identified in Liangshan Black Sheep,Hu Sheep,Bamei Sheep,and Poll Dorset.Genotype visualization analysis indicated that the frequency of homozygous wild-type KIT allele in Liangshan Black Sheep was significantly higher than in white breeds.Additionally,the frequencies of homozygous mutant alleles the TMEM18,TRPM1,MC1R,and ASIP genes in Liangshan Black Sheep were also significantly higher than in white sheep breeds.【Conclusion】Our findings indicated that the KIT,MC1R,ASIP,TMEM18,and TRPM1 are key genes for wool color formation in Liangshan Black Sheep.This research provides a theoretical basis for a deeper understanding of the genetic mechanisms underlying wool coloration in these sheep.
