摘要
目的建立冬瓜皮药材的高效液相色谱法(HPLC)特征图谱,并同时测定其牡荆素鼠李糖苷、异牡荆素-2″-O-鼠李糖苷等黄酮类成分的质量分数,结合聚类分析(cluster analysis,CA)、主成分分析(principal component analysis,PCA)及正交偏最小二乘判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)分析数据,对不同产地的冬瓜皮进行整体评价。方法采用Dikma Platisil ODS(250 mm×4.6 mm,5μm)色谱柱,以乙腈(A)和0.1%甲酸(B)为流动相梯度洗脱,检测波长为270 nm,流速为1.0 mL/min,柱温为35℃。以黄酮类含量、特征图谱共有峰相似度为主要评价指标,结合化学模式分析其不同产地冬瓜皮药材的差异。结果15批冬瓜皮样品特征图谱均呈现5个共有特征峰,与对照图谱相似度均大于0.900,指认出4-羟基苯甲酸、牡荆素鼠李糖苷、异牡荆素-2″-O-鼠李糖苷3种成分,并对黄酮类成分进行定量测定。聚类分析聚为4类,与PCA分析结果一致,通过VIP值筛选出主成分物质牡荆素鼠李糖苷为影响分类的差异性物质,OPLS-DA也进一步验证了上述结果。结论该HPLC特征图谱及含量测定方法稳定可靠,可用于评价不同产地冬瓜皮的质量,为冬瓜皮药材质量控制及其冬瓜皮药用开发提供科学依据。
Objective To establish a high-performance liquid chromatography(HPLC)characteristic chromatogram method for winter melon peel medicinal materials,and simultaneously determine the contents of flavonoid components such as vitexin rhubarb and isovitexin-2″-O-rhubarb.Combined with the analysis data of cluster analysis(Cluster Analysis),principal component analysis(Principal Component Analysis),and orthogonal partial least squares discriminant analysis(Orthogonal Partial Least Squares Discriminant Analysis),an overall evaluation of winter melon peels from different origins was conducted.Methods A Dikma Platisil ODS(250 mm×4.6 mm,5μm)chromatographic column was used.Acetonitrile(A)and 0.1% formic acid(B)were used as the mobile phases.Gradient elution was performed.The detection wavelength was 270 nm,the flow rate was 1.0 mL/min,and the column temperature was 35℃.Taking the flavonoid content and the similarity of common peaks in characteristic spectra as the main evaluation indicators,combined with the chemical model,the differences of winter melon peel medicinal materials from different origins were analyzed.Results The characteristic spectra of the 15 batches of winter melon peel samples all presented 5 common characteristic peaks,and the similarity with the control spectra was all greater than 0.900.Three components,namely 4-hydroxybenzoic acid,vitexin rhamnoside,and isovitexin-2″-O-rhamnoside,were identified,and the flavonoid components were quantitatively determined.Cluster analysis classified them into 4 categories,which was consistent with the results of PCA analysis.Through VIP values,the principal component substance vitrecin rhunoside was screened out as the differential substance affecting the classification.Orthogonal partial least squares discriminant analysis also further verified the above results.Conclusion The characteristic chromatograph(HPLC)and content determination method are stable and reliable,which can be used to evaluate the quality of winter melon peels from different origins,providing a scientific basis for the quality control of winter melon peel medicinal materials and the medicinal development of winter melon peels.
