摘要
以黄河三角洲野生灰绿碱蓬(Suaeda glauca)根际土壤样品为试验材料,富集和分离出能够代谢菲(Phenan-threne,Phe)的碱蓬根际细菌,通过分析菌株的溶磷能力和在不同盐度下降解Phe的性能,筛选出兼具植物促生和耐盐能力的高效Phe降解菌,盆栽实验检验该菌对碱蓬生长的实际促生效果和土壤中Phe的降解能力。结果显示,以Phe为唯一碳源初筛获得的25株Phe降解菌中,有3株菌能够同时溶解有机磷和无机磷,将这3株菌分别接种于添加了不同浓度NaCl的含Phe培养基中,7天后菌株sg-517能够在添加3%NaCl的盐环境中实现32.48%的Phe降解率,证明该菌为兼具促生和耐盐性能的Phe降解菌;向Phe污染土壤中接种菌株sg-517,利用盆栽实验证实了该菌株能够同时提高碱蓬幼苗的生长指标和土壤中Phe的去除效率,实现了提高碱蓬植株抗逆性和促进Phe代谢/吸收的双重效果;利用16S rDNA和BLAST分析,菌株sg-517鉴定为羽扇豆布鲁氏菌(Brucella lupini)。本研究结果有助于获取多功能微生物资源和促进植物-微生物联合修复技术的开发应用。
Rhizosphere soil of wild Suaeda glauca from coastal wetland of the Yellow River Delta was used to screen phenanthrene(Phe)degrading bacteria.By analyzing the performance of phosphorus solubilization and Phe degradation of different salinity levels,the effective PH-degrading bacteria with both plant growth promo-tion and salt tolerance were screened out,and the actual growth promotion effect and degradation ability of Phe in soil were tested by pot experiment.The results showed that among the 25 strains obtained from the prelimi-nary screening,3 strains could dissolve both organophosphates and inorganic phosphorus.Then these 3 strains were inoculated in the medium containing Phe and different concentrations of NaCl.After 7 days,strain sg-517 showed 32.48%degradation of Phe in the medium under 3%NaCl treatment.Therefore,strain sg-517 was proved potentials of Phe degradation,plant growth promotion and salt-tolerance.After inoculation of strain sg-517 into Phe contaminated soil,both growth indexes of suadea seedlings and removal efficiency of Phe were improved,which confirmed the practical effect of strain sg-517 on promoting the stress resistance of plants and PAHs metabolism/absorption.By 16S rDNA sequencing and BLAST analysis,the strain sg-517 was identi-fied as Brucella lupini.The results of this study help to obtain multifunctional microbial resources and promote the development and application of bioremediation technology combined by plant and microorganism.
