摘要
藻毒素在全球海域广泛分布,严重危害生态系统与人体健康。氮杂螺环酸(Azaspiracids, AZAs)是一种含氮且具独特螺环结构的聚醚类藻毒素,可引起人体腹泻、呕吐等症状。AZAs的来源主要是Azadinium属和Amphidoma属,是制备AZAs标准物质的主要来源。本文采用批次培养和单因素分析的方法分别探讨不同温度、盐度和光照强度条件下腹孔环胺藻(Azadinium poporum)AZDY06生长和产毒的状况,探究腹孔环胺藻生长和产毒的最佳培养条件。结果表明,温度为22℃,盐度为26,光照强度为80μmol·m^(-2)·s^(-1)时腹孔环胺藻生长良好,AZA-2产量最高。在指数生长期胞内毒素占比高,稳定生长期毒素逐渐释放到胞外,且稳定生长期单位细胞毒素产量大于对数生长期,温度是影响毒素产量的最重要变量,其次是光照强度。本研究可为腹孔环胺藻的培养提供技术参数,以获得更多的AZA-2毒素,为毒素的分离纯化提供来源。
Phycotoxins are widely distributed in marine environments worldwide,which seriously threaten the health of ecosystem and human health.Azaspiracids(AZAs)are a kind of nitrogen-containing polyether phycotoxins with unique spirocyclic structures,which can cause diarrhea,vomiting,and other symptoms for the poisoned consumers.Microalgae Azadinium and Amphidoma are the main producers of AZAs,which could be used for preparation of reference materials of AZAs.Batch culture and univariate analysis of temperature,salinity and light intensity were carried out to investigate optimal conditions for the growth and toxin production of A.poporum AZDY06 in this study.The results showed that AZDY06 grew well and produced the highest amount of AZA-2 at the temperature of 22℃,salinity of 26 and light intensity of 80μmol·m^(-2)·s^(-1).The proportion of intracellular toxin was high during the exponential growth period,and AZA-2 was gradually released into the extracellular during the stable growth period,and the toxin production per cell was more in the stable phase than that in the exponential growth period,and the temperature was the most important variable influencing the toxin production,followed by the light intensity.The results of this study can provide technical parameters for the culture of A.poporum to obtain more AZA-2 toxin and provide a source for toxin isolation and purification.
