摘要
为建立半红树植物苦槛蓝组织快繁体系,以幼嫩腋芽作为外植体,采用正交设计等试验方法,研究不同消毒时间、不同类型培养基和不同浓度外源激素等因素对苦槛蓝愈伤组织诱导、不定芽诱导和生根的影响。结果显示:(1)最佳消毒时间为0.1%HgCl2消毒3 min;(2)愈伤组织诱导最佳培养基配方为MS培养基+0.6 mg·L-16-BA+0.06 mg·L-1 NAA+30 g·L-1蔗糖+5.5 g·L-1琼脂,pH值为6.2;(3)不定芽继代培养基配方为MS培养基+0.6 mg·L-16-BA+0.03 mg·L-1 NAA+30 g·L-1蔗糖+5.5 g·L-1琼脂,pH值为6.2;(4)生根诱导培养基配方为1/2 MS培养基+0.3 mg·L-1 NAA+20 g·L-1蔗糖+6.5 g·L-1琼脂,pH值为6.2;(5)将草炭土、蛭石和河沙按照3∶1∶1比例混匀制备成移栽基质,育苗成活率可达到86%。
To establish a rapid in vitro propagation system for mangrove associate species Pentacoelium bontioides,the young axillary bud was used as an external implant to study the effects of different disinfection time,different types of culture medium and different concentrations of exogenous hormones on induction of callus,adventitious buds and rooting.The results showed that:(1)the optimum disinfection method was 0.1%HgCl2 treated for 3 min;(2)The optimum medi⁃um for callus induction was MS medium+0.6 mg·L-16-BA+0.06 mg·L-1 NAA+30 g·L-1 sucrose+5.5 g·L-1 agar,pH 6.2;(3)The optimum medium for the induction of adventitious bud was MS medium+0.6 mg·L-16-BA+0.03 mg·L-1 NAA+30 g·L-1 sucrose+5.5 g·L-1 agar,pH 6.2;(4)The optimum medium for rooting formula was 1/2 MS medium+0.3 mg·L-1 NAA+20 g·L-1 sucrose+6.5 g·L-1 agar,pH 6.2;(5)Seedling survival rate could reach 86%when it was transplanted into substrate consisted of peat soil,vermiculite,and river sand with a ratio of 3∶1∶1.
