摘要
目的探讨熄风解郁方对帕金森抑郁(depression in Parkinson′s disease,dPD)大鼠的作用机制。方法采用腹腔注射脂多糖(lipopolysaccharide,LPS)4 d建立dPD大鼠模型。采用行为学检测评价抑郁样行为和运动能力,脑部磁共振成像技术检测脑区自发性活动和白质纤维完整性,免疫组化法检测c-Fos、离子钙结合蛋白1(anti-allograft inflammatory factor 1,Iba-1)及酪氨酸羟化酶(tyrosine hydroxylase,TH)抗体表达,Western blot法检测Notch信号通路关键分子蛋白表达,实时荧光定量聚合酶链反应检测Notch信号通路关键分子mRNA表达。结果熄风解郁方可提高dPD大鼠的糖水偏好率、旷场实验的水平运动总距离及中心区活动距离、转棒实验的停留时间;改善脑区自发性活动及白质纤维损伤;增加c-Fos表达及黑质区TH表达,降低Iba-1表达;降低Notch信号通路关键分子的mRNA及蛋白表达。结论熄风解郁方可能通过抑制Notch信号通路表达,减轻神经炎症,发挥治疗dPD的作用。
Objective To investigate the mechanism of Xifeng Jieyu Prescription(熄风解郁方,XJP)in rats with depression in Parkinson′s disease(dPD).Methods A rat model with dPD was established by lipopolysaccharide(LPS)intraperitoneal injection for 4 days.The spontaneous activity of brain regions and the integrity of white matter fibers were detected by magnetic resonance imaging.The expressions of c-Fos,Iba-1 and TH antibodies were detected by immunohistochemistry.The protein expression of Notch signaling pathway was detected by western blot technology.The mRNA expression of Notch signaling pathway was detected by quantitative real-time polymerase chain reaction.Results XJP increased the sucrose preference,horizontal total distance,central distance and standing time of rotarod test in rats with dPD,improved the spontaneous brain activity and white matter fiber damage,increased the expressions of c-Fos and TH,and decreased the expression of Iba-1.The mRNA and protein expressions of Notch signaling pathway were decreased.Conclusion XJP may reduce neuroinflammation and plays a role in the treatment of dPD by inhibiting the gene and protein expression of Notch signaling pathway.
