基于RNA-seq探究光周期对扬州鹅睾丸发育的影响机制

摘要 旨在探究光周期对扬州鹅睾丸发育的影响及机制。试验对象为经产扬州鹅(735 d,公母比例为1∶5),饲养于全封闭人工控制光照和温度的棚舍内(1500只/棚),饲养期间光照程序为短光照(L∶D=6.5 h∶17.5 h)饲养60 d,随后延长光照(L∶D=11.5 h∶12.5 h)饲养,分别于短光照第38天(短光照组)以及长光照第73天(长光照组)采集血液,分离血清并检测相关的生理生化指标和激素水平;采集其左侧睾丸组织进行转录组测序(RNA sequencing,RNA-seq)分析,筛选不同光周期的差异基因,并对差异基因进行富集分析。结果:与短光照组相比,长光照组睾丸体积增大、曲细精管中有大量的成熟精子,且曲细精管直径、上皮高度、管腔直径、精子数量均有显著升高(P<0.05);长光照组血液中睾酮(T)、促卵泡激素(FSH)、黄体生成素(LH)浓度均显著高于短光照组(P<0.05);RNA-seq测序结果显示,与短光照组相比,长光照组扬州鹅睾丸组织中表达上调的基因有4383个,表达下调的基因有2928个;其差异表达基因主要富集于细胞外基质(ECM)与细胞表面受体之间的相互作用、磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)信号通路、核糖体等通路。综上,延长光照后PI3K-AKT通路被激活,从而促进扬州鹅睾丸发育,为深入认识光照对动物繁殖的调控作用提供了参考资料。 This study aimed to explore the mechanism of the effect of photoperiod on the development of goose testis.The experimental sub⁃jects were Yangzhou geese(735 days of age,male to female ratio of 1∶5),raised in fully enclosed artificial sheds with controlled light and temperature(1500 geese per shed).During the feeding period,the light program was short light(L∶D=6.5 h∶17.5 h)feeding for 60 days,followed by extended light(L∶D=11.5 h∶12.5 h)feeding.The blood samples were collected from the geese on the 38th day of short light(the short light group)and the 73rd day of long light(the long light group),and serum was separated to detect relevant physio⁃logical and biochemical indicators and hormone levels.The left testicular tissue samples were used for transcriptome sequencing(RNA seq)analysis;and differentially expressed genes(DEGs)were screened between the short light and long light groups.The results showed that the testis volume was increased in the long light group,compared with that in the short light group,together with a large number of mature sperm in the seminiferous,increased seminiferous tubule diameter,epithelial height,and lumen diameter(P<0.05).The levels of blood testoster⁃one(T),follicle-stimulating hormone(FSH),and luteinizing hormone(LH)concentrations were significantly higher in the long light group than those in the short light group.The transcriptome sequencing results showed that 4383 genes were significantly upregulated and 2928 genes were downregulated in the long light group,compared with those in the short light group.The DEGs were mainly enriched in ECM-receptor interaction,the PI3K-AKT signaling pathway,Ribosome,and so on.It can be concluded that the PI3K-AKT pathway was activated by prolonged light,thereby promoting the development of goose testis.This experiment provided important evidence for further un⁃derstanding the regulatory mechanism of light on animal reproduction.
