摘要
为探究万寿竹属植物染色体特征,采用常规制片技术,对长蕊万寿竹、金佛山万寿竹及独山万寿竹的染色体进行核型分析。结果表明,3种植物根尖以0.05%秋水仙素和0.002 mol/L 8-羟基喹啉等体积混合液4℃处理1.5 h,卡诺氏固定液4℃固定24 h,无水乙醇和浓盐酸等体积混合液常温解离9 min,改良石炭酸品红染色液常温染色15 min为宜;长蕊万寿竹、金佛山万寿竹及独山万寿竹的核型公式分别为2 n=2 x=16=12 sm+4 st(1SAT)、2 n=2 x=16=2 m+12 sm+2 st、2 n=2 x=16=10 sm+6 st(1SAT)。染色体相对长度均为2 n=16=4 L+4 M2+2 M1+6 S构成,核型均属于3B型。根据核型不对称性分析,3种植物中长蕊万寿竹进化程度最高,金佛山万寿竹最低。
To explore the chromosomal characteristics of the plants of Disporum species,the karyotype analysis of the chromosomes of Disporum longistylum,Disporum jinfoshanense and Disporum dushanense was conducted using conventional preparation techniques.The results showed that the root tips of the three plants were treated with a mixed solution of 0.05%colchicine and 0.002 mol/L 8-hydroxyquinoline in equal volumes at 4℃for 1.5 hours,and fixed with Carnot's fixative at 4℃for 24 hours.The mixed solution of equal volumes of anhydrous ethanol and concentrated hydrochloric acid was dissociated at room temperature for 9 minutes,and the modified carbolic acid and fuchsin staining solution was suitable for staining at room temperature for 15 minutes.The karyotype formulas for Disporum longistylum,Disporum jinfoshanense and Disporum dushanense were classified as 2 n=2 x=16=12 sm+4 st(1SAT),2 n=2 x=16=2 m+12 sm+2 st,and 2 n=2 x=16=10 sm+6 st(1SAT).The relative lengths of the chromosomes were all composed of 2 n=16=4 L+4 M2+2 M1+6 S,and the karyotypes all belonged to type 3B.According to the analysis of karyotype asymmetry,among the three plants,the Disporum longistylum had the highest degree of evolution,while the Disporum jinfoshanense had the lowest.
