组培技术促进狭叶薰衣草繁殖及分子育种

摘要 本研究以狭叶薰衣草叶片及带芽茎段为外植体,以MS为基本培养基,研究了灭菌方式、2,4-D、6-BA、NAA不同激素浓度组合对其愈伤组织、芽诱导以及增殖和生根的影响。结果表明:外植体最佳灭菌方式为,15%次氯酸钠灭菌12 min,污染率为19.3%;最佳不定芽诱导培养基MS激素配比为1.0 mg/L 6-BA与0.1mg/L NAA组合,诱导率86.67%;最佳生根培养基MS激素配比为0.1mg/L NAA,生根率最高为80%,根系均匀。狭叶薰衣草的组织培养,可以满足快繁技术的要求,繁殖系数高、同时保留母株的优良性状。 In this study,the leaves and stem segments with buds of Lavandula angustifolia Mill.were used as explants and MS was used as the basic medium to study the effects of sterilization methods,different hormone concentrations of 2,4-D,6-BA and NAA on callus,bud induction,proliferation and rooting.The results showed that the best sterilization method of explants was 15%sodium hypochlorite sterilization for 12 min,and the pollution rate was 19.3%.The best adventitious bud induction medium MS hormone ratio was 1.0 mg/L 6-BA and 0.1 mg/L NAA combination,the induction rate was 86.67%,the best rooting medium MS hormone ratio was 0.1 mg/L NAA,the rooting rate was 80%,the root system was uniform.The tissue culture of Lavandula angustifolia Mill.can satisfy the requirements of rapid propagation technology,with high propagation coefficient and retaining the good characters of the parent plants.
