摘要
目的:探讨肾气丸及其拆方对糖尿病肾脏病(diabetic kidney disease,DKD)小鼠肾功能与水通道蛋白(aquaporins,AQPs)的影响。方法:将24只db/db小鼠随机分为模型组(Mo组)、肾气丸组(S-R组,10.4 g·kg^(-1))、肾气丸-补肾组(S-T组,8.1 g·kg^(-1))及肾气丸-利尿组(S-D组,2.3 g·kg^(-1)),每组6只;另取6只db/m小鼠作为对照组(C组)。取小鼠尾尖血检测空腹血糖(fasting blood glucose,FBG)、糖化血清蛋白(glycosylated serum protein,GSP)水平。采用ELISA法检测血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)、血肌酐(serun creatinine,SCr)、尿素氮(blood urea nitrogen,BUN)、微量白蛋白(microalbumin,mAlB)、尿肌酐(urine creatinine,UCr)水平以及24小时尿蛋白定量(24 hour urinary protein,UP)。HE染色法检测肾脏皮质组织病理形态,Western blot法检测肾脏组织AQP1蛋白表达水平。结果:与C组比较,Mo组小鼠FBG水平升高(P<0.01);与Mo组比较,S-D组小鼠FBG水平降低(P<0.01)。与C组比较,Mo组小鼠GSP水平升高(P<0.01);与Mo组比较,S-R组、S-D组小鼠GSP水平降低(P<0.05)。与C组比较,Mo组小鼠血清TC、TG、LDL-C水平升高,HDL-C水平降低(P<0.05);与Mo组比较,S-R组、S-T组小鼠血清TC、TG、LDL-C水平降低,HDL-C水平升高(P<0.05)。与C组比较,Mo组小鼠SCr、BUN、mAlB、UP水平升高,UCr水平降低(P<0.01);与Mo组比较,S-D组小鼠SCr、BUN、mAlB、UCr、UP水平降低(P<0.05);与Mo组比较,S-R、S-T组小鼠SCr、BUN、mAlB、UP水平降低(P<0.05)。HE染色显示,C组小鼠肾皮质无明显病理变化;Mo组小鼠肾皮质中肾小球系膜增生,伴基质沉积;S-R组、S-T组、S-D组小鼠仅见皮质区肾小球系膜增生。与C组比较,Mo组小鼠肾脏AQP1蛋白表达水平降低(P<0.01);与Mo组比较,S-R组小鼠肾脏AQP1蛋白表达水平升高(P<0.05)。结论:肾气丸及其拆方均能改善DKD小鼠的肾损伤,其机制可能与调节AQP1蛋白表达有关。
Objective:To investigate the effects of Shenqi Pill and its modified formula on renal function and aquaporins(AQPs)in diabetic kidney disease(DKD)in mice.Methods:Twenty-four db/db mice were randomly divided into the model group(Mo group),Shenqi Pill group(S-R group,10.4 g·kg^(-1)),Shenqi Pill-kidney tonic group(S-T group,8.1 g·kg^(-1)),and Shenqi Pill-diuretic group(S-D group,2.3 g·kg^(-1)),with six mice in each group;and six db/m mice were taken as the control group(C group).Fasting blood glucose(FBG)and glycosylated serum protein(GSP)levels were measured by taking the tail tip blood of mice.Serum total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),and triglyceride(TG)were measured by ELISA.density lipoprotein cholesterol(HDL-C),serun creatinine(SCr),blood urea nitrogen(BUN),microalbumin(mAlB),urine creatinine(UCr),and urinary creatinine(UCr).The levels of serun creatinine(SCr),urea nitrogen(bloodurea nitrogen,BUN),microalbumin(mAlB),urinary creatinine(UCr),and 24 hour urinary protein(UP)were quantified at 24 h.The pathological morphology of renal cortical tissues was detected by HE staining method,and the protein expression level of AQP1 in renal tissues was detected by Western Blot method.Results:Compared with group C,the FBG level of mice in Mo group was increased(P<0.01);compared with Mo group,the FBG level of mice in S-D group was decreased(P<0.01).Compared with group C,the level of GSP in mice in Mo group was increased(P<0.01);compared with Mo group,the level of GSP in mice in S-R and S-D groups was decreased(P<0.05).Compared with group C,the serum TC,TG,LDL-C levels of mice in Mo group were increased,and HDL-C levels were decreased(P<0.05);compared with Mo group,the serum TC,TG,LDL-C levels of mice in S-R group and S-T group were decreased,and HDL-C levels were increased(P<0.05).Compared with group C,mice in Mo group had increased levels of SCr,BUN,mAlB,and UP,and decreased levels of UCr(P<0.01);mice in S-D group had decreased levels of SCr,BUN,mAlB,UCr,and UP compared with Mo group(P<0.05);and mice in S-R and S-T groups had decreased levels of SCr,BUN,mAlB,and UP compared with Mo group(P<0.05).HE staining showed that there were no obvious pathological changes in the renal cortex of mice in group C;glomerular thylakoid hyperplasia in the renal cortex of mice in group Mo,accompanied by matrix deposition;and glomerular thylakoid hyperplasia in the cortical area only was seen in mice in groups S-R,S-T,and S-D.Compared with group C,the renal AQP1 protein expression level of mice in Mo group was reduced(P<0.01);compared with Mo group,the renal AQP1 protein expression level of mice in S-R group was increased(P<0.05).Conclusion:Shenqi Pill and its modified formula can improve renal injury in DKD mice,and its mechanism may be related to the regulation of AQP1 protein expression.
