摘要
【目的】滩羊成无根状同源异型盒-4(ALX4)基因组织表达和多态性及其与羊毛性状的关联性,为滩羊裘皮分子标记辅助选育提供依据。【方法】针对前期研究中筛选出的位点采用SNP分型技术和RT-PCR技术对107只滩羊进行ALX4基因多态性检测,并与初生期和二毛期羊毛性状进行关联分析。【结果】在滩羊ALX4基因上筛选出3个SNPs位点,分别为g.72453265 T>C、g.72455864 T>C和g.72476712 T>C,其中g.72453265 T>C位点产生TT、TC和CC 3种基因型,g.72455864 T>C位点产生TT、TC 2种基因型,g.72476712 T>C位点产生TT、TC 2种基因型;基因多态性分析表明,3个SNPs位点属于低度多态,SNP1位点均处于非Hardy-Weinberg平衡状态(P<0.05),SNP2和SNP3位点均处于Hardy-Weinberg平衡状态(P>0.05);关联分析结果显示:SNP1Chr15:72453265位点CC基因型出生侧部弯曲数显著大于其他基因型(P<0.05),SNP3 chr15:72476712位点TT基因型二毛侧部毛长显著大于其他基因型(P<0.05);组织表达分析表明,出生时期滩羊皮肤组织中ALX4基因的表达量极显著高于二毛期滩羊的皮肤组织(P<0.01)。【结论】ALX4基因多态性与滩羊羊毛性状显著相关,可作为滩羊羊毛性状选育的候选基因,其中Chr15:72453265位点和chr15:72476712位点可能是影响滩羊羊毛弯曲和毛长的潜在候选SNP位点,可作为滩羊羊毛性状的遗传分子标记。
【Objective】The study aimed to investigate the tissue expression and polymorphisms of the aristaless-like homeobox-4(ALX4)gene in Tan sheep,as well as their association with wool traits,to provide a basis for molecular marker-assisted breeding of Tan sheep pelt quality.【Method】Based on previously screened loci,SNP genotyping and RT-PCR techniques were employed to detect the polymorphism of ALX4 gene in 107 Tan sheep,and the correlation were conducted between ALX4 gene polymorphism and wool traits at birth and second wool stage.【Result】Three SNPs were identified in the ALX4 gene of Tan sheep,which were g.72453265 T>C,g.72455864 T>C,and g.72476712 T>C.respectively.The g.72453265 T>C locus exhibited three genotypes(TT,TC,CC),while the other two loci each had two(TT,TC).Polymorphism analysis showed that the three SNPs were low polymorphic,and the SNP1 locus was in non-Hardy-Weinberg equilibrium(P<0.05).The results of association analysis showed that the CC genotype of SNP1 Chr15:72453265 locus had a significantly greater number of lateral curvatures than other genotypes(P<0.05),and the TT genotype of SNP3 chr15:72476712 locus had a significantly greater length of lateral hairs than other genotypes(P<0.05).Tissue expression analysis showed that the expression of ALX4 gene in the skin tissue of Tan sheep at birth was significantly higher than that in the skin tissue of Tan sheep at second wool stage(P<0.01).【Conclusion】The polymorphism of ALX4 gene is significantly correlated with the wool traits of Tan sheep,which can be used as a candidate gene for the breeding of wool traits in Tan sheep.Chr15:72453265 locus and chr15:72476712 locus may be potential candidate SNP loci affecting wool bending and wool length of Tan sheep,which can be used as genetic molecular markers for wool traits of Tan sheep.
