摘要
[目的]分析虱螨脲对草地贪夜蛾[Spodoptera frugiperda(Smith)]幼虫生长发育及其肠道细菌多样性的影响,探究该害虫对虱螨脲胁迫的响应机制,为合理利用虱螨脲防治草地贪夜蛾及其抗药性机理研究提供科学依据。[方法]采用浸叶法测定10%虱螨脲悬浮剂对草地贪夜蛾3龄幼虫的毒力,并确定该药剂对草地贪夜蛾3龄幼虫的致死中浓度(LC_(50));运用分离培养法分别对虱螨脲LC_(50)处理的处理组和吐温水处理的对照组草地贪夜蛾3龄幼虫进行肠道可培养细菌分离,根据菌株菌落形态及16S rDNA序列对分离获得的可培养细菌进行种属鉴定。[结果]虱螨脲对草地贪夜蛾3龄幼虫表现出较强的毒力,且其毒力随着处理浓度的增加和处理时间的延长而增强,接触药剂后96 h的LC_(50)为1.162 mg/L。从处理组草地贪夜蛾3龄幼虫肠道分离到可培养细菌9株,分属于4门6科6属,从对照组分离到可培养细菌13株,分属于4门7科8属。在门分类水平上,虱螨脲LC_(50)处理后,厚壁菌门(Firmicutes)、变形菌门(Proteobacte-ria)、放线菌门(Actinobacteria)的相对丰度分别较对照组降低了10.15%、6.01%、0.09%,而芽孢杆菌门(Bccillota)的相对丰度增加了16.24%;在属分类水平上,肠球菌属(Enterococcus)、肠杆菌属(Enterobacter)、葡萄球菌属(Staphylococ-cus)的相对丰度较对照组分别下降了6.64%、4.65%、2.14%,而芽孢杆菌属(Bacillus)的相对丰度增加了16.24%。[结论]虱螨脲对草地贪夜蛾幼虫肠道微生物群落组成、结构与多样性具有明显影响。
[Objective]The effects of lufenuron on the growth and development of Spodoptera frugiperda(Smith)lar-vae and the diversity of intestinal bacteria were analyzed,and the response mechanism of Spodoptera frugiperda(Smith)to lufenuron stress was explored,which provided scientific basis for the rational usage of lufenuron to control Spodoptera frugiperda(Smith)and its resistance mechanism.[Method]The toxicity of 10%lufenuron suspension to the 3rd instar lar-vae of Spodoptera frugiperda(Smith)was determined by leaf dipping method,and the lethal medium concentration(LC_(50))on the 3rd instar larvae of Spodoptera frugiperda(Smith)was determined.Intestinal culturable bacteria of 3rd instar larvae of Spodoptera frugiperda(Smith)in lufenuron LC_(50) treatment group and Tween-20 treatment control group were isolated by isolation culture method,and the species of the isolated culturable bacteria were identified according to the colony morphology and 16S rDNA sequence.[Result]Lufenuron showed strong toxicity against the 3rd instar larvae of Spodoptera frugiperda(Smith),and its toxicity increased with the increase of concentration and treatment time.The LC_(50) was 1.162 mg/L at 96 h after exposure to agent.Nine strains of culturable bacteria were isolated from the intestine in 3rd in-star larvae of lufenuron LC_(50) treatment group,belonging to 6 genera in 6 families under 4 phyla,while the control group had 13 strains belonging to 8 genera in 7 families under 4 phyla.At the phylum level,after lufenuron LC_(50) treatment,the relative abundance of Firmicutes,Proteobacteria and Actinobacteria decreased by 10.15%,6.01%and 0.09%respectively compared to control group,and the relative abundance of Bacillota increased by 16.24%.At the genus level,the relative abundance of Enterococcus,Enterobacter and Staphylococcus decreased by 6.64%,4.65%and 2.14%respectively com-pared with the control,and the relative abundance of Bacillus increased by 16.24%.[Conclusion]Lufenuron has great effects on the structure,function and diversity of its intestinal flora of Spodoptera frugiperda(Smith).
