摘要
【目的】进一步了解刺梨原浆对大肠杆菌的抗菌作用机制。【方法】采用微量稀释法测定不同浓度刺梨原浆对大肠杆菌的最小抑菌浓度、对大肠杆菌生长曲线的影响、对大肠杆菌生物被膜形成的作用以及大肠杆菌的耐药谱。分析刺梨原浆对大肠杆菌生长、增殖及耐药表型的影响。利用RNA-seq和qRT-PCR对刺梨原浆胁迫下大肠杆菌转录组和差异表达基因进行分析,阐明刺梨原浆对大肠杆菌生物学特性的调控作用。【结果】刺梨原浆对多重耐药大肠杆菌具有明显的抗菌作用,最小抑菌浓度为12.50%的刺梨原浆。随着刺梨原浆作用浓度的增加和作用时间的延长,12 h后大肠杆菌的生长繁殖受到明显抑制。刺梨原浆对大肠杆菌生物被膜的形成存在抑制作用,其中6.25%的刺梨原浆对大肠杆菌生物被膜形成的抑制率达47%。此外,刺梨原浆作用下,能够逆转大肠杆菌对β-内酰胺类、氨基糖苷类和四环素类抗生素的耐药性。刺梨原浆胁迫下,大肠杆菌中与鞭毛运动、代谢过程、抗生素生物合成以及生物膜形成等相关基因出现差异表达,如编码细菌趋化性的基因tsr显著下调,其在大肠杆菌鞭毛运动、代谢抗生素的生物合成方面发挥作用,编码细胞运动和黏附相关的基因pdeH显著下调,其在控制生物被膜生长的过程中发挥作用。【结论】刺梨原浆具有潜在的抗菌增敏活性,为系统阐明刺梨原浆的抗菌机理提供理论支撑,也为刺梨原浆的应用拓宽途径。
[Objective]This study aimed to explore the antibacterial activity and mechanism of action of Rosa roxburghii Tratt juice on Escherichia coli.[Methods]In this study,the minimum inhibitory concentration(MIC),growth curve,biofilm formation,and antibiotic susceptibility of E.coli treated with Rosa roxburghii Tratt juice were determined to analyze the effects of Rosa roxburghii Tratt juice on the growth,proliferation,and resistance phenotypes of E.coli.RNA-seq and qRT-PCR were used to analyze the transcriptome and differentially expressed genes of E.coli under Rosa roxburghii Tratt juice stress,elucidating the regulatory role of Rosa roxburghii Tratt juice.[Results]Rosa roxburghii Tratt juice exhibited significant antibacterial activity against multidrug-resistant E.coli,with a minimum inhibitory concentration of 12.50%.As the concentration of Rosa roxburghii Tratt juice increased and the exposure time prolonged,the growth and proliferation of E.coli were significantly inhibited after 12 hours.Rosa roxburghii Tratt juice inhibited the formation of biofilms in E.coli,with a 47%inhibition rate observed with 6.25%Rosa roxburghii Tratt juice.In addition,the application of Rosa roxburghii Tratt juice could reversed the resistance of E.coli to β-lactam,aminoglycosides and tetracyclines.Under Rosa roxburghii Tratt juice stress,E.coli showed differential expression of genes related to flagellar movement,metabolic processes,antibiotic biosynthesis,and biofilm formation.Genes encoding bacterial chemotaxis such as tsr were significantly downregulated,playing a role in flagellar movement and antibiotic biosynthesis in E.coli,while genes like pdeH,involved in cell motility and adhesion,were significantly downregulated,influencing the control of biofilm growth.[Conclusion]This study demonstrates that Rosa roxburghii Tratt juice has potential antibacterial sensitizing activity,providing theoretical support for elucidating its antibacterial mechanism and expanding the application pathways of Rosa roxburghii Tratt juice.
