依据生长性状和简单重复序列标记的红松二代优树联合选择

Integrated Selection of the Second-Generation Elite Trees of Pinus koraiensis Based on Growth Traits and Simple Sequence Repeat Markers

摘要 为了评选红松(Pinus koraiensis)二代优树,为营建红松二代种子园提供理论和材料支持,以辽宁地区1989年营建的红松优树半同胞子代测定林为研究对象,通过调查参试单株的树高和胸径,开展各性状的差异性分析,并估算遗传参数,采用选择指数法评选候选优树,结合简单重复序列分子标记技术,对该群体的遗传多样性和亲缘关系进行分析,通过不同的取样方法筛选出红松二代优树,进而构建指纹图谱。结果表明:生长性状在家系间、区组间的差异均达到显著或极显著水平,且存在较为丰富的变异,表型变异系数范围6.66%~29.80%。树高、胸径、单株材积的家系遗传力及单株遗传力分别为0.46、0.29、0.35和0.07、0.34、0.22,除树高的单株遗传力较低外,其余性状的2类遗传力均受中度遗传控制。采用选择指数法评选出51株候选优树,单株材积的平均遗传增益和现实增益分别为21.36%和97.08%。候选优树群体的遗传多样性水平较高,平均有效等位基因数、多态性信息含量、香农信息指数、观测杂合度、期望杂合度分别为2.397、0.455、0.978、0.484、0.511。该候选群体存在着不同程度的亲缘关系,通过聚类分析,可划分为8个组。采用Power Core version的不同方法对该候选群体进行取样,M策略的遗传参数水平较高,接近于候选群体,且材积增益高于候选群体,因此,最终入选的红松二代优树为17株,总入选率为4%。同时,在12对多态性引物中筛选出3对引物构建了17株红松二代优树的指纹图谱。 In order to select the second-generation elite trees of Pinus koraiensis and provide theoretical and material support for the establishment of the second-generation seed orchard of P.koraiensis,a half-sib progeny test forest of elite trees of P.koraiensis established in Liaoning region in 1989 was taken as the research object.By investigating the tree height and diameter at breast height(DBH)of the tested individual trees,the difference analysis of various traits was carried out,and the genetic parameters were estimated.The selection index method was used to select the candidate elite trees.Combined with the simple sequence repeat(SSR)molecular marker technology,the genetic diversity and genetic relationship of this population were analyzed.The second-generation elite trees of P.koraiensis were screened out through different sampling methods,and then the fingerprinting was constructed.The results showed that the differences in growth traits among families and blocks reached significant or extremely significant levels,and there was relatively rich variation,with the phenotypic variation coefficient ranging from 6.66%to 29.80%.The family heritabilities and individual heritabilities of tree height,DBH,and individual volume were 0.46,0.29,0.35 and 0.07,0.34,0.22 respectively.Except for the relatively low individual heritability of tree height,the two types of heritabilities of the remaining traits were moderately genetically controlled.51 candidate elite trees were selected by the selection index method,and the average genetic gain and realized gain of individual volume were 21.36%and 97.08%respectively.The candidate elite tree population had a relatively high level of genetic diversity,with the average number of effective alleles,polymorphism information content,Shannon information index,observed heterozygosity,and expected heterozygosity being 2.397,0.455,0.978,0.484,and 0.511 respectively.There were different degrees of genetic relationships in this candidate population.Through cluster analysis,it could be divided into 8 groups.Different methods of Power Core version were used to sample this candidate population.The genetic parameter level of the M strategy was relatively high,close to that of the candidate population,and the volume gain was higher than that of the candidate population.Therefore,17 second-generation elite trees of P.koraiensis were finally selected,with a total selection rate of 4%.At the same time,3 pairs of primers were screened out from 12 pairs of polymorphic primers to construct the fingerprinting of 17 second-generation elite trees of P.koraiensis.
