不同干燥工艺牡丹皮提取物高效液相指纹图谱建立及其抗氧化作用研究 被引量:1

Development of HPLC Fingerprint and Evaluation of Antioxidant Activity in Moutan Cortex Extracts Processed by Different Drying Methods

摘要 目的:研究不同干燥工艺对牡丹皮提取物中丹皮酚、芍药苷的含量及其抗氧化作用的影响。方法:采用减压干燥、烘干和冷冻干燥对4个批次牡丹皮水提液进行干燥;高效液相色谱(HPLC)法测定各提取物中丹皮酚和芍药苷含量并建立指纹图谱;测定3种干燥工艺下牡丹皮提取物的2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)和1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率。结果:3种干燥工艺下牡丹皮提取物中丹皮酚含量为0.0188~1.4789μg/mL,且含量从高到低依次为冷冻干燥>烘干>减压干燥;芍药苷含量为0.7301~1.2129μg/mL。12个批次样品的相似度为0.812~0.973;不同干燥工艺下提取物(质量浓度1000μg/mL)的ABTS自由基清除率为52.45%~78.64%,且冷冻干燥>烘干>减压干燥;不同干燥工艺对提取物(质量浓度1000μg/mL)的DPPH自由基清除率为51.48%~64.37%。结论:不同干燥工艺对牡丹皮提取物中丹皮酚含量和ABTS自由基清除能力影响较大。采用冷冻干燥法替代传统干燥工艺能减少丹皮酚含量损失,提高抗氧化能力,可为牡丹皮及其制剂开发提供参考。 Objective:To investigate the effects of different drying methods on the content of paeonol and paeoniflorin in Moutan Cortex extracts and its antioxidant activity.Methods:Four batches of aqueous extracts of Moutan Cortex were subjected to vacuum drying,oven drying,and freeze drying.The content of paeonol and paeoniflorin in each extract was determined using high-performance liquid chromatography(HPLC),and fingerprint analysis was established.The antioxidant activity of the extracts obtained from the three drying methods was evaluated based on their 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)and 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging capacities.Results:The paeonol content in the extracts ranged from 0.0188 to 1.4789μg/mL,with the highest content observed in freeze-dried samples,followed by oven drying and vacuum drying.The paeoniflorin content ranged from 0.7301 to 1.2129μg/mL.The sample similarity in the 12 batches ranged from 0.812 to 0.973.At a concentration of 1000μg/mL,the ABTS radical scavenging rate of the extracts ranged from 52.45%to 78.64%,following the order freeze drying>oven drying>vacuum drying.The DPPH radical scavenging rate ranged from 51.48%to 64.37%across different drying methods.Conclusion:Different drying methods significantly affect the paeonol content and ABTS radical scavenging capacity of Moutan Cortex extracts.Freeze drying is recommended as an alternative to traditional drying methods,as it reduces paeonol loss and enhances antioxidant activity,providing a reference for the development of Moutan Cortex and its preparations.
