摘要
建立糖尿病肾病合并骨质疏松大鼠模型,分为模型组、L-六味地黄丸组(0.75 g/kg)、M-六味地黄丸组(1.5 g/kg)、H-六味地黄丸组(3.0 g/kg)、H-六味地黄丸+RANKL重组蛋白(H-六味地黄丸+RANKL组)(3.0 g/kg六味地黄丸+2.0 mg/kg RANKL重组蛋白),正常组给予普通饲料以及饮用自来水.随后对大鼠进行糖尿病及骨代谢相关指标检测;ELISA法检测IL-6、IL-1β、TNF-α水平;HE检测股骨组织病理学变化.Western blot检测RANKL/RANK/OPG通路相关蛋白表达.探讨六味地黄丸调节RANKL/RANK/OPG信号通路对糖尿病肾病合并骨质疏松大鼠的作用及机制.结果显示与正常组相比,模型组骨小梁稀疏、间隙变大、数量减少,骨髓腔增大,FBG、HOMAIR、TG、TC、IL-6、IL-1β、TNF-α水平、肾肥大指数及RANKL、RANK蛋白表达升高,FINS、血磷、血钙水平及OPG蛋白表达降低(P<0.05);与模型组比较,L-六味地黄丸组、M-六味地黄丸组、H-六味地黄丸组FBG、HOMA-IR、TG、TC、IL-6、IL-1β、TNF-α水平、肾肥大指数及RANKL、RANK蛋白表达降低,骨小梁数量、FINS、血磷、血钙水平及OPG蛋白表达升高,不同剂量六味地黄丸组间存在统计学差异(P<0.05);与H-六味地黄丸组比较,H-六味地黄丸+RANKL组FBG、HOMA-IR、TG、TC、IL-6、IL-1β、TNF-α水平、肾肥大指数及RANKL、RANK蛋白表达升高,骨小梁数量、FINS、血磷、血钙水平及OPG蛋白表达降低(P<0.05).表明六味地黄丸通过调节RANKL/RANK/OPG信号通路改善糖尿病肾病合并骨质疏松大鼠症状.
The purpose of this study was to explore the effect and mechanism of Liuwei Dihuang Pill diabetes nephropathy with osteoporosis rats,and its effect on RANKL/RANK/OPG signaling pathway.In this study,the rat model of diabetes nephropathy with osteoporosis was established,and was divided into model group,L-Liuwei Dihuang Pill group(0.75 g/kg),M-Liuwei Dihuang Pill group(1.5 g/kg),H-Liuwei Dihuang Pill group(3.0 g/kg),H-Liuwei Dihuang Pill+RANKL protein(3.0 g/kg Liuwei Dihuang Pill+2.0 mg/kg RANKL recombinant protein).The normal group was given normal diet and tap water.Then,the indexes related to diabetes and bone metabolism were detected in rats.ELISA was used to measure IL-6,IL-1β,and TNF-α.HE was used to detect pathological changes in femoral tissue,and western blot was used to detect the expression of proteins related to RANKL/RANK/OPG pathway.The results showed that compared with normal group,model group had sparse bone trabeculae,larger gaps,reduced quantity,enlarged bone marrow cavity,the FBG,HOMA-IR,TG,TC,IL-6,IL-1β,TNF-α levels,renal hypertrophy index,and RANKL and RANK protein expression were higher,while FINS,blood phosphorus,blood calcium,and OPG protein expression were lower(P<0.05).Compared with model group,the FBG,HOMA-IR,TG,TC,IL-6,IL-1β,TNF-α levels,renal hypertrophy index,and RANKL and RANK protein expression were lower in the L-Liuwei Dihuang Pill group,M-Liuwei Dihuang Pill group,and H-Liuwei Dihuang Pill group,while the number of bone trabeculae,FINS,blood phosphorus,blood calcium,and OPG protein expression were higher,there were statistically prominent differences between different doses of Liuwei Dihuang Pill groups(P<0.05).Compared with the H-Liuwei Dihuang Pill group,the FBG,HOMA-IR,TG,TC,IL-6,IL-1β,TNF-α levels,renal hypertrophy index,and RANKL and RANK protein expression were higher in H-Liuwei Dihuang Pill+RANKL protein,while the number of bone trabeculae,FINS,blood phosphorus,blood calcium,and OPG protein expression were lower(P<0.05).In conclusion,Liuwei Dihuang Pill can improve the symptoms of diabetes nephropathy with osteoporosis rats by regulating RANKL/RANK/OPG signaling pathway.
