油菜脂肪酸合成相关基因2-酮脂酰-CoA还原酶克隆及生物信息学分析

Gene Clone and Bioinformatics Analysis of 2-Ketoacyl-CoA Reductase Gene Which Related to Fatty Acid Synthesis of Brassica napus

摘要 油酸是菜籽油主要脂肪酸,有很好的营养保健功能,菜籽油品质改良对改善我国居民营养水平十分重要,因而受到油菜育种研究者的广泛关注,但其合成的分子机理研究仍有待于进一步提高。为促进我国油菜产业发展,以高油酸油菜5叶期-6叶期叶片为材料,克隆了油酸代谢相关基因2-酮酯酰-CoA还原酶,同时还构建了过表达载体,测序分析发现基因长度为1212 bp,生物信息学分析发现该基因编码312个蛋白,通过Predictprotein定位于内质网,生物信息学分析表明,该序列与数据库公布序列同源性为99%,CDs序列和氨基酸序列分析结果都表明,甘蓝型油菜KCR与白菜型油菜KCR聚类在一起,拟南芥KCR与亚麻荠KCR聚类在一起,甘蓝型油菜KCR与白菜型油菜KCR亲缘关系较近,同拟南芥KCR、亚麻荠KCR亲缘关系相对较远。 Rapeseed is the largest self-produced source of vegetable oil in China,accounting for 15%of the total consumption of edible oil in China.Improving the quality of rapeseed oil is very important for improving the nutritional level of Chinese residents,and has received widespread attention from rapeseed breeding researchers.Oleic acid is the main fatty acid in rapeseed oil and has good nutritional and health functions,but the molecular mechanism of its synthesis still needs further improvement.This study used high oleic acid rapeseed leaves at the 5-6 leaf stage as materials to clone the oleic acid metabolism related gene 3-ketoacyl CoA reductase.At the same time,an overexpression vector was also constructed,and sequencing analysis revealed a gene length of 1212 bp.Bioinformatics analysis revealed that the gene encodes 312 proteins,which are located in the endoplasmic reticulum through Predictprotein.The sequence has a 99%homology with the publicly available sequence in the database.The analysis of CDs and amino acid sequences showed that the KCR of Brassica napus and Brassica napus were clustered together,while the KCR of Arabidopsis thaliana was clustered together with the KCR of Shepherd’s purse.The KCR of Brassica napus was closely related to the KCR of Brassica napus,but relatively distant from the KCR of Arabidopsis thaliana and Shepherd’s purse.
