玫瑰中药复方对紫外线诱导HaCaT细胞损伤的修复作用

摘要 目的研究玫瑰中药复方对紫外线(UV)诱导HaCaT细胞损伤的修复作用。方法用UV诱导HaCaT细胞损伤模型;采用CCK8法检测玫瑰中药复方对HaCaT正常细胞和损伤细胞的影响;DPPH法和ABTS+法检测玫瑰中药复方的自由基清除能力;β-半乳糖苷酶染色法检测玫瑰中药复方对UV诱导HaCaT细胞损伤的保护作用;流式细胞术检测玫瑰中药复方对细胞凋亡的影响;采用试剂盒检测细胞内丙二醛(MDA)、过氧化氢酶(CAT)含量和超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)活性;采用试剂盒检测玫瑰中药复方处理后细胞内活性氧(ROS)的水平变化;RT-qPCR检测细胞中p16和p21基因的表达水平。结果结果显示玫瑰中药复方具有体外抗氧化能力,同时对UV诱导的HaCaT损伤细胞具有抑制细胞凋亡和促增殖的作用,并通过增强抗氧化酶(CAT、SOD、GPx)活性来降低细胞内ROS水平,降低氧化产物MDA水平,增强损伤细胞ABTS+和DPPH自由基清除能力,以及显著降低损伤细胞内氧化损伤标志基因p16和p21 mRNA水平。结论玫瑰中药复方对UV损伤的细胞具有修复作用。 Objective To determine the repair effect of a rose herbal compound on ultraviolet(UV)-induced damage in HaCaT cells.Methods The UV-induced HaCaT cell damage model was established.The effect of the rose herbal compound on both normal and damaged HaCaT cells was assessed with CCK8 method.The DPPH and ABTS+assays were used to evaluate the compound free radical scavenging capabilities.β-galactosidase staining was used to examine the protective effect of rose herbal compound against UV-induced HaCaT cell damage.Flow cytometry was used to assess the rose herbal compound impact on the cell apoptosis.Commercial kits were used to measure intracellular malondialdehyde(MDA),catalase(CAT),superoxide dismutase(SOD),and glutathione peroxidase(GPx)levels after the treatment with rose herbal compound.Changes in reactive oxygen species(ROS)levels inside the cells after the treatment were also evaluated with the commercial kit.RT-qPCR was performed to analyze the expression levels of the p16 and p21 genes in the cells.Results Rose herbal compound displayed obvious in vitro antioxidant capabilities.It not only inhibited the apoptosis and promoted the proliferation in UV-induced damaged HaCaT cells,but also reduced intracellular ROS levels by enhancing the activities of antioxidant enzymes(CAT,SOD,and GPx).Furthermore,it decreased MDA levels,enhanced the ability to clear ABTS+and DPPH free radicals in damaged cells,and significantly lowered the mRNA levels of oxidative damage markers p16 and p21.Conclusion Rose herbal compound helps repaire UV-damaged cells.
