摘要
目的探讨大黄灵仙胶囊对大鼠炎性胆管组织中miRNA-152-3p、miRNA-221-3p及miRNA-30b的表达水平及临床意义。方法运用随机数字表法将30只SD大鼠分为假手术组、模型组及治疗组,每组10只。假手术组采用麻醉后开腹再予缝合进行造模,模型组、治疗组使用3 mg/kg的脂多糖(lipopolysaccharide,LPS)进行胆总管注射构建胆管炎性大鼠模型。造模后假手术组、模型组予2 mL/100 g体质量蒸馏水灌胃,每日2次,每次间隔12 h;治疗组予320 mg/(kg·d)的大黄仙灵胶囊颗粒溶液在造模前2 d开始灌胃给药至造模后3 d,每日2次,每次间隔12 h。采用实时定量PCR法检测大鼠胆管组织中miRNA-152-3p、miRNA-221-3p及miRNA-30b的表达水平。结果与假手术组比较,模型组miRNA-152-3p表达水平较低(P<0.001)、miRNA-221-3p表达水平较低(P<0.001)及miRNA-30b表达水平较低(P<0.01),差异具有统计学意义。与模型组比较,治疗组miRNA-152-3p表达水平较高(P<0.001)、miRNA-221-3p表达水平较高(P<0.05)、miRNA-30b表达水平较高(P<0.01),差异具有统计学意义。结论大黄灵仙胶囊具有保护胆管组织的作用,作用机制可能与调控miRNA-152-3p、miRNA-221-3p及miRNA-30b表达,从抗炎、抗氧化应激角度发挥保护胆管的作用。
Objective To investigate the influence of Dahuang Lingxian Capsule(大黄灵仙胶囊)on expression levels of miRNA-152-3p,miRNA-221-3p and miRNA-30b in in flammatory bile duct tissues of the rats and the clinical significance.Methods A random number table method was used to divide 30 SD rats into sham surgery group,model group and treatment group,with 10 rats in each group.The sham group used abdomen opening after anesthesia and then suturing for modeling,and the model group and treatment group used 3 mg/kg lipopolysaccharide(LPS)for common bile duct injection to construct a rat model of cholangitic inflammation.After modeling,the sham surgery group and the model group were given 2 mL/100 g body weight distilled water by gavage,twice a day,with an interval of 12 hours each time.The treatment group was given 320 mg/(kg·d)solution of Dahuang Lingxian Capsule,and gastric administration was started 2 days before modeling,and administered to 3 days after modeling,twice a day,with an interval of 12 hours each time.The expression levels of miRNA-152-3p,miRNA-221-3p and miRNA-30b in rat bile duct tissues were detected by real-time quantitative PCR.Results Compared with that of the sham group,the miRNA-152-3p expression level was lower(P<0.001),the expressions of miRNA-221-3p(P<0.001)and miRNA-30b(P<0.01)were significantly lower in the model group.Compared with the model group,the treatment group had higher expressions of miRNA-152-3p(P<0.001),miRNA-221-3p(P<0.05)and miRNA-30b(P<0.01),and the differences were statistically significant.Conclusion Dahuang Lingxian Capsule has the effect of protecting bile duct tissue,and the mechanism of action may be related to regulating the expressions of miRNA-152-3p,miRNA-221-3p and miRNA-30b,and play a role in protecting bile ducts from the perspective of anti-inflammatory and oxidative stress.
