光及补氮诱导对三角褐指藻岩藻黄素合成积累的影响

摘要 该文探究了光及补氮诱导培养模式对三角褐指藻岩藻黄素合成积累的影响.研究结果显示,在光照色温4000 K、光照周期21 h(光照)∶3 h(黑暗)、初始光照强度4000 lux诱导培养192 h后光照强度降至3000 lux条件下,获得的三角褐指藻岩藻黄素含量和产量分别为13.67 mg·g^(-1)和44.93 mg·L^(-1),分别为未进行光诱导培养模式的1.31倍和1.77倍.在此最佳光诱导条件下,流加补氮培养进一步提升了三角褐指藻生物量、岩藻黄素含量和产量,分别达到4.46 g·L^(-1)、15.77 mg·g^(-1)和70.62 mg·L^(-1),较未进行光及补氮诱导培养模式分别提高了1.89倍、1.51倍和2.78倍.实验证实了光优化及流加补氮培养可有效促进三角褐指藻生物量和岩藻黄素积累,且培养获得的岩藻黄素微生物和重金属含量均符合GB 26405—2011标准.研究结果为利用三角褐指藻规模化生产岩藻黄素及应用提供了新思路. The effects of light and nitrogen supplementation-induced culture mode on the synthesis and accumulation of fucoxanthin of Phaeodactylum tricornutum(P.tricornutum)were investigated.The study showed that under a light color temperature of 4000 K,a photoperiod of 21 h(light)∶3 h(dark),and an initial light intensity of 4000 lux induced incubation for 192 h followed by a decrease in light intensity to 3000 lux,the content and yield of 13.67 mg·g^(-1)and 44.93 mg·L^(-1)of fucoxanthin in P.tricornutum were obtained,which were 1.31 and 1.77 times higher than the culture mode without light-induced,respectively.The flow-added supplemental tryptone fermentation culture under this optimal light-induced condition further enhanced the biomass,fucoxanthin content and yield of P.tricornutum,which reached 4.46 g·L^(-1),15.77 mg·g^(-1)and 70.62 mg·L^(-1),respectively,which were 1.89,1.51 and 2.78 times higher than the culture mode without light and nitrogen supplementation-induced.It was confirmed that light optimization and flow-added nitrogen supplementation culture could effectively promote the biomass and fucoxanthin accumulation of P.tricornutum,and the microbial and heavy metal content of fucoxanthin obtained from the culture complied with the standard of GB 26405-2011.The results of the study provide a new idea for the large-scale production of fucoxanthin and its application using P.tricornutum.
