摘要
为研究豆蔻明(CDN)对慢性热应激儋州鸡肝脏氧化损伤的影响及潜在机制,本试验将200只儋州鸡随机分为5组,分别为对照组(CON),热应激组(HS),低剂量豆蔻明组(L-CDN),中剂量豆蔻明组(M-CDN),高剂量豆蔻明组(H-CDN)。CON组在(25±2)℃条件下饲养,其余组温度控制在(36±2)℃。CON组和HS组饲喂基础日粮,L-CDN、M-CDN、H-CDN组分别在基础饲粮中添加50、100、200 mg/kg CDN。试验期42 d,预试期21 d,正试期21 d。42日龄从每个重复选取2只体重接近的儋州鸡屠宰。HE(Hematoxylin-Eosin Staining)染色观察结果表明:与CON组相比,HS组造成肝脏水泡变性,肝细胞索排列紊乱。同CON组相比,HS组丙二醛(MDA)含量提高51.72%,超氧化物歧化酶(SOD)含量降低42.82%(P<0.05)、过氧化氢酶(CAT)含量降低20.53%(P<0.05)、谷胱甘肽过氧化物酶(GSH-Px)活性水平降低17.23%(P<0.05)。同HS组相比,H-CDN组GSH-Px、SOD含量分别提高15.29%、64.41%(P<0.05),MDA含量降低21.53%(P<0.05)。此外,与HS组相比,H-CDN组促炎因子白细胞介素(IL-1β)水平降低27.39%(P<0.05),核转录因子(NF-κB)的表达量降低19.33%(P<0.05),抗炎因子白介素(IL-10)的表达量提高64.74%(P<0.05),改善了热应激儋州鸡氧化还原状态。综上,饲喂豆蔻明能够提高儋州鸡肝脏抗氧化能力,缓解热应激诱导的氧化应激和炎症反应,对肝脏有一定保护作用,且饲粮中添加200 mg/kg CDN的效果最好。
The purpose of this study was to investigate the effect of cardamonn(CDN)on oxidative damage in the liver of Danzhou chickens under chronic heat stress and its potential mechanism.A total of 200 Danzhou chickens were randomly divided into 5 groups,namely control group(CON),heat stress group(HS),low-dose cardamonin group(L-CDN),medium-dose cardamonin group(M-CDN),high-dose cardamonin group(H-CDN),CON group were raised at(25±2)°C,and the temperature of the test group was controlled at(36±2)°C.The CON group and HS group were fed basal diet,and 50,100,200 mg/kg CDN were added to the basal diet with L-CDN,M-CDN and H-CDN groups,respectively.The test period was 42 days,the pre-test period was 21 days,and the main test period was 21 days.At 42 days of age,2 Danzhou chickens with similar body weight were selected from each replicate for slaughter.The results of histopathological observation showed that compared with the CON group,the HS group caused liver vesicular degeneration and hepatocyte cord arrangement.Compared with the CON group,the content of malondialdehyde(MDA)in the HS group increased by 51.72%,the content of superoxide dismutase(SOD)decreased by 42.82%(P<0.05),the content of catalase(CAT)decreased by 20.53%(P<0.05),and glutathione peroxidase(GSH-Px)activity level decreased by 17.23%(P<0.05).Compared with the HS group,the content of GSH-Px,SOD in the H-CDN group increased by 15.29%and 64.41%(P<0.05),respectively,and the content of MDA decreased by 21.53%(P<0.05).In addition,compared with the HS group,the level of pro-inflammatory factor interleukin(IL-1β)in the H-CDN group decreased by 27.39%(P<0.05),the expression level of nuclear transcription factor(NF-κB)decreased by 19.33%(P<0.05),and the expression level of anti-inflammatory factor interleukin(IL-10)increased by 64.74%(P<0.05),which improved the redox state of heat-stressed Danzhou chickens.The above results suggested that feeding cardamine could improve the antioxidant capacity of Danzhou chicken liver,alleviate the oxidative stress and inflammatory response induced by heat stress,and had a certain protective effect on the liver,and the addition of 200 mg/kg CDN to the diet had the best effect.
